Cryptococcus neoformans has been studied systematically in China. Ecologically, natural isolates from pigeon droppings were found in the polymorphism of phenotypes which includes serotype A, AD and urease negative strains of C. neoformans var. neoformans, Those phenotypes also were found correspondingly in clinical isolates. In molecular biology, G+C mol% and karyotypes of C. neoformans were analysed. A significant information got from PFGE analysis was that two varieties and five serotypes of C. neoformans isolates were shown obvious different patterns of karyotypes. Clinically, a new medium was made for the clinical identification of this pathogenic yeast, which is favorable to test both urease and phenoloxidase simultaneously. The L-form of C. neoformans was induced by a high osmotic culture medium. It suggests that the chronic course and clinical relapses of infection may correlate with the L - form formation during the infectious periods of this yeast. Fluconazole has been shown very efficient to cure the patients infected by C. neoformans. A possible combination therapy , such as fluconazole and amphotericin B liposome, should be emphasized in the future research. In conclussion, the systemic study on C. neoformans has been preliminary carried out in China and some interesting results have been achieved.

Using this primer, of 24 clinical and environmental isolates of serotype A of C.neoformans and 8 standard strains investigated by PCR, 20 strains produced complete identical fingerprinting patterns, the other 4 strains had different fingerprinting patterns. 2 strains of serotype B and C yielded indistinguishable fingerprinting patterns. Conclusions ①The majority of strains of serotype A had similar and stable fingerprinting patterns. ②Some strains of serotype B and C had an indistinguishable fingerprinting patterns. ③The same serotype strains from different sources may produce different DNA fingerprinting patterns. ④The DNA fingerprinting is a rapid, simple and feasible method for identifying Cryptococcus neoformans.

目的 用随机引物扩增新生隐球菌临床和环境分离株的DNA ,观察DNA指纹与血清型的关系 ,探索新生隐球菌基因分型标准。方法 采用 3种不同寡核苷酸重复序列 [CN1(GTG) 5、CN2(GACA) 4和CN3(GATA) 4]为引物 ,对从临床和鸽粪中分离到的 2 4株血清A型和 8株四种血清型的新生隐球菌标准株的DNA进行扩增 ,根据扩增产物的电泳带型来对比DNA指纹和血清型之间的关系。结果  2 4株临床和鸽粪分离的血清A型株中 ,2 0株产生和标准株完全相似的DNA带型 ,1株与标准D型DNA带型相似。 3株的PCR指纹与血清A、B、C、D型均不相同。 2株B型和 2株C型产生几乎完全一致的DNA指纹带型。结论 ①大多数血清A型新生隐球菌菌株具有相似DNA指纹带型。②血清B型和C型 (gattii变种 )的DNA带型不易区别。③相同血清型不同的株可能有不同的DNA指纹带型。④PCR法用于新生隐球菌菌株鉴定是一种快速、方便、可行的方法。

Thirty two Cryptococcus neoformans strains were isolated from 144 samples of pigeon droppings in private and public square pigeon shelters in Shanghai on GAEC. The positive isolation rate was 22.2% (32/144). All strains are Cry. neoformans var. neoformans and serotype A identified by Crypto. Cheek Kit(Iatron laboratories Inc Tokyo, Japan). Also, Christensen's Urea testes were all positive. Mice were inoculated by two strains in abdomen and same strains can be isolated again. All strains can grow well at 37℃...

采用GAEC培养基培养了144份私人和公共养鸽场所的鸽粪标本,从中共分离得32株新生隐球菌,阳性率达22.2%。37℃生长良好。尿素试验均为阳性。血清因子分型结果证实阳性标本均为新生隐球菌新生变种,全是A血清型,动物接种后能重新分离出该菌。病理证实具致病性。
