酞丁安是我国在临床使用的抗病毒化疗药物之一,为了优化其活性,增加其水(脂)溶性,对酞丁安结构中氨基侧链部分及母体进行结构修饰与改造,共合成十个酞丁安类似物,其抗病毒作用表明化合物IIIc,VIIa.与VIIb,显示抗疱疹病毒活性,其中化合物IIIc活性最强,其体外抗病毒活性较酞丁安强10倍,其余化合物均无活性。国家自然科学基金资助课题

METHODS The pharmacokinetics of TCPN and the tissue distribution of TCPN was studied with radioreceptor assay and radioactive isotope assay respectively. RESULTS The concentration time curve after sc 50 μg·kg -1 of TCPN was fitted to first order absorption two compartment model: with 2 28 h of T 12β 0 125 h of T max , 3 44 μg·L -1 of C max and 23 0 L·kg -1 ·h -1 of Cl. After sc 50 μg·kg -1 of TCPN,the gradient of peak radioactivity in the tissues was kidney>liver>brain>submandibular glands>colon>heart>skeletal muscle. As compared with the other tissues, the redioactivity in brain remained at a higher level with a longer eliminating time and obviously decreased after sc QNB. CONCLUSIONS That TCPN eliminated rapidly in blood but slowly in brain was related to the binding of TCPN to muscarinic acetylcholine receptors in brain.

AIM To determine whether Ganoderma polysaccharide influenced the expression levels of IL 2 and IL 3 mRNA in murine splenocytes in vitro. METHODS Reverse transcription polymerase chain reaction(RT PCR) and gel documentation analysis system were exploited to detect and semi quantify the mRNA expression levels. RESULTS Ganoderma polysaccharide, GLB 7, (5,10,20 and 40 mg·L -1 ) was added to murine primary splenocyte culture at the beginning of incubation and the levels of mRNA expressions for IL 2 and IL 3 in the cells were assayed after 12 h or 21 h incubation to determine if any concentration effect relationship exists. It was found that the levels of IL 2 and IL 3 increased in a concentration dependent manner with GLB 7 within limited concentrations. The increase rates for IL 2 mRNA were 10 4%, 21 9%, 37 8% and 46 6% respectively, and for IL 3 mRNA,15 4%, 35 2%, 52 4% and 74 5% respectively. The activities of IL 2 and IL 3 in the supernatants were also enhanced markedly in GLB 7 treated cultures compared with the untreated ones with a limited concentration dependent manner. CONCLUSION The immunopotentiating effect of Ganoderma polysaccharide may be associated with its potentiation of IL 2 and IL 3 mRNA expressions at the transcriptive level.

目的观察灵芝多糖体外对小鼠脾细胞IL-2、IL-3mRNA的表达水平是否有影响。方法逆转录多聚酶链式反应(RT-PCR)检测mRNA表达,凝胶图象光密度分析系统进行相对定量。结果在原代小鼠脾细胞培养开始时加入不同浓度的灵芝多糖GLB7(5,10,20,40mg·L-1),培养12h及21h后观察GLB7与IL-2及IL-3mRNA表达水平之间的量效关系。发现在一定范围内,随着GLB7浓度的升高,IL-2及IL-3mRNA表达水平也相应提高,对IL-2mRNA的提高率分别为10.4%,21.9%,37.8%,46.6%;对IL-3mRNA的提高率分别为15.4%,35.2%,52.4%,74.5%。1998-04-08收稿,1998-07-26修回*国家自然科学基金资助课题,No39400165作者简介:王庆彪,男,26岁,硕士;雷林生,男,38岁,医学博士,副教授培养12h和30h的上清液中IL-2样活性及IL-3样活性处理组与对照组相比亦有明显升高,且有一定的浓度依赖关系。结论灵芝多糖的免疫增强作用与其在转录水平上促进IL-2及IL-3mRNA的表达有关
