We first and foremost used the micronucleus test to rapidly evaluate the effects of cystamine and H2O2 administered separately or in combination, on radiation induced chromosomal damage in bone-marrow cells and Ehrish ascites tumour cells. We first and foremost used the micronucleus test to rapidly evaluate the effects of cystamine and H2O2 administered separately or in combination, on radiation induced chromosomal damage in bone-marrow cells and Ehrish ascites tumour cells. The results of chromosomal damage examined by micronucleus test are as follow:1. Tumor bearing mice are administered with cystamine either orally or intrape-ritoneally, and then exposed to double or single dose irradiation. The radioprotective effect of cystamine on bone-marrow cells is significant, but not significant on Ehrish ascites tumor cells.2. H2O2 can increase the radiosensitivity of chromosomal damage in Ehrish ascites tumor cells, but has no effect in bone-marrow cells.3. When cystamine and H2O2 are simultaneously administered to tumor-bearing mice, H2O2 does not decrease the radioprotective effect of cystamine on bone-marrow cells, but the cystamine can decrease the radiosensitive effect of H2O2 on Ehrish ascites tumor cells.

本研究工作首次应用微核测定技术快速评价胱胺和H_2O_2在照射前分别或合并使用时,它们对带瘤小鼠的骨髓细胞和艾氏腹水癌细胞染色体辐射损伤的影响。微核测定的结果表明: (1)照前口服或腹注胱胺对受一次或分次照射的腹水癌小鼠骨髓细胞的染色体损伤均有显著性的保护作用,而对其癌细胞的染色体损伤无明显的防护效果。(2)H_2O_2能够提高腹水癌细胞染色体辐射损伤的敏感性,而对其骨髓细胞的染色体损伤无明显的影响。(3)胱胺和H_2O_2合并使用时,H_2O_2不降低胱胺对骨髓细胞的防护作用,而胱胺却能削弱H_2O_2对癌细胞的致敏作用。

Normal mouse bone marrow cell suspensions sealed in diffusion chambers were implanted intraperitoneally into normal mice and mice rendered neutropenia by pretreatment with vincristine or irradiation with 750 rad ~(60)Co. After five days' 'intraperitoneal incubation the diffusion chambers were removed and the number of colonies was scored. Our experiments showed that mouse bone marrow colonies were able to grow in unirradiated hosts as well as in hosts treated with irradiation or cytotoxic drugs. The samples...

Normal mouse bone marrow cell suspensions sealed in diffusion chambers were implanted intraperitoneally into normal mice and mice rendered neutropenia by pretreatment with vincristine or irradiation with 750 rad ~(60)Co. After five days' 'intraperitoneal incubation the diffusion chambers were removed and the number of colonies was scored. Our experiments showed that mouse bone marrow colonies were able to grow in unirradiated hosts as well as in hosts treated with irradiation or cytotoxic drugs. The samples assayed in our experiments yielded about 80~90 colonies per 10~5 cells cultured. There were no significant differences in the number of colonies among these three groups. Some reports suggested that serum from the animals irradiated or pretreated with cytotoxic drugs contained a higher CSF activity, but this has not been demonstrated in the present report. Probably, the endogenous CSF activity in normal mouse used in over experiments was sufficient to stimulate the colony formation of mouse bone marrow CFU-C.

本文报道了采用不同寄主小鼠进行小鼠造血干细胞体内扩散盒琼脂培养的结果比较。寄主小鼠在植入扩散盒1~2小时前接受750拉德~(60)Co照射或在植入前24小时注射VCR1.6毫克/公斤并未能提高CFU-C细胞团产率。这种情况是否是由于正常小鼠体液内的CSF水平已足以有效地刺激小鼠骨髓CFU-C生成细胞团,故提高CSF浓度不能再增加细胞团的产率。有待在掌握本实验小鼠的种属特性及调整照射剂量后作进一步研究。

This paper reports the relative inhibitory specificity and possible biochemical mechanisms of the anticancer effect of Disodium Cantharidate (DSC). The results showed that, in a concentration of 25μg/ml, DSC inhibited 49% of the ~3H-TdR incorporation in Ehrlich Ascites Carcinoma (EAC), while in bone marrow cells only 16%. Furthermore, DSC in the same concentration inhibited 70% of the ~3H-Leu incorporation in EAC, while in bone marrow cells only 34%. The preliminary results indicated that DSC could have some...

本文报告斑蝥酸钠抗癌作用的生化机理与相对的抑癌专一性。体外实验结果表明:当斑蝥酸钠浓度为25微克/毫升时,可抑制艾氏腹水癌细胞~3H-胸苷参入的49%,而对骨髓细胞的~3H-胸苷参入仅抑制16%;同样的斑蝥酸钠浓度,抑制艾氏腹水癌细胞的~3H-亮氨酸参入的70%,而对骨髓细胞仅抑制34%。上述二者的差别均有显著性。 荧光偏振度测定的初步实验结果还表明:斑蝥酸钠能影响癌细胞膜的流动性及胞浆的结构性。
