In the present paper some factors affecting the superinduction of interferon in diploid cells were studied. However, the duration of treatment with inhibitors of nucleic acid synthesis ( mithramycin and actinomy-cin D ) should not exceed one hour before the termination of superinduction. The. behaviour of superinduction by mithramycin was similar to that by actinomycin D The optimal time for combined poly I : C and cycloheximide treatment was about 9 hours. Under our experimental conditions, the interferon yield in the superinduction system was increased by 170-248 fold as compared with similar system without superinduction. The present paper describes a procedure for the large scale production of fibroblast interferon.

Two functional parameters evaluated by NBT dye reduction and YC rosette formation and a morphological change assessed by Wright-Giemsa stain of HL-60 cells were used to determine the extent of HL-60 cell differentiation induced by retinoids and DMSO. Studies revealed that under the action of retinoic acid(RA) and its two new synthetic analogs, 4-(ethoxycarbophenyl) retiamide(RI) and 4-(hydroxycarbophenyl) retiamide (RII), HL-60 cells could be induced to differentiate to more mature cells with functional characteristics of granuloeytes. When HL-60 cells were incubated with 10~(-6)M of RⅠ and RⅡ for 6 days, 42.3% and 43.6% of the eells were capable of reducing NBT; 20.7% and 14.9% could form YC rosettes with yeast ceils covered by human C_3 complement. DMSO was also shown to be active in the induction of both functional differentiation phenotypes. However, the expression of C_3 complement receptor was much earlier than the emergence of NBT reduction capacity under its induction. It is resonable to speculate that the induction machanism of retinoids and DMSO is somewhat different. Both retinoids and DMSO could induce HL-60 cells into morphologically more mature myeloid cells including myeloeytes, metamyelocytes and banded neutrophils. However, only a few cells matured into fully segmented ones. The induction of differentiation of HL-60 cells by all of the drugs tested was found to be associated with a reduction of the number of viable ceils. Since this phenomenon appeared later than cell differentiation, it may be related with the change of growth characteristics of differentiated cells.

本文研究了新维甲酸衙生物——维胺酯(RⅠ)和维胺酸(RⅡ)对人早幼粒白血病细胞(HL-60)的分化诱导作用,并与维甲酸(RA)和二甲基亚砜(DMSO)进行了比较。HL-60细胞经RⅠ和RⅡ处理后,细胞对NBT染料还原能力增强,细胞表面出现C_3补体受体等中性粒细胞的分化特征,但作用比RA弱。DMSO诱导上述分化性状出现时间不同,提示维甲酸类药物与DMSO诱导分化的机制可能不同。上述药物均可使HL-60细胞形态按粒系途径向较成熟的方向发展,但变化出现较晚。

We studied the effects of sodium butyrate on inducingearly antigen of EBV(EA) and enhancing the transformation of lymphocytes by EBV. The results showed that sodium but yrate not only induces alone or cooperates with tumor promoter-12 - 0-tetradecanoyl-phorbol-13-acetate(TPA) to induce EBV-EA. but also enhances or cooperates with TPA to enhance the transformation of lymphocytes by EBV. We studied the effects of sodium butyrate on inducingearly antigen of EBV(EA) and enhancing the transformation of lymphocytes by EBV. The results showed that sodium but yrate not only induces alone or cooperates with tumor promoter-12 - 0-tetradecanoyl-phorbol-13-acetate(TPA) to induce EBV-EA. but also enhances or cooperates with TPA to enhance the transformation of lymphocytes by EBV. The positive cells of EBV-EA is 0.4% at the concentration of 0.4mM/ml. with the concentration of sodium butyrate increasing, the positive cells of EBV-EA increased. The positive cells of EBV EA is 25% at the concentration of 40mM/ml. But sodium butyrate has some toxic effects. The differences of the numbers of colonies between experimental and EBV control group are not significant at the concentration of 0.1mM/ml. The number of the colonies of experimental group is 2.9 times as high as that of EBV control group. And the number of colonies of experimental group cooperation with TPA is 5.1 times as high as that of EBV control group.

本文研究丁酸钠对EB病毒的诱导作用及对EB病毒转化淋巴细胞的促进作用。通过实验,证明丁酸钠不仅能诱导或协同促癌物TPA诱导EB病毒的早期抗原,而且还能促进或协同促癌物质TPA促进EB病毒对淋巴细胞的转化。
