In this experiment, Disseminated Intravascular Coagulation(DIC) model was Prepared on New Zealand rabbits to observe the change of cytosolic calcium concentration [Ca2 +]i of thrombocytes measured with Quin-2 , in order to study the significance of [Ca2 +]i of thrombocytes in the occurance of endo-toxin-induced DIC. The effect of endotoxin on [Ca2+]i of platelets in vitro was also studied. The results indicated- the resting cytosolic calcium concentration of platelets of New Zealand rabbits is 112±24 nM (n= 7 ) . In vitro endotoxin can directly increase [Ca2+]i of platelet and also this effect is dose-dependent. In vivo, the experimental DIC models were prepared on New Zealand rabbits and the [Ca2+]i of platelets increase from 112 ±24 nM in normal to 343 ±48 nM, The results suggest that endotoxin can increase of [Ca2+]i of platelet in vitro and in vivo. In DIC model the platelets maintain a high level of [Ca2 +]i and result in the "hypercoagulant" of platelet, which may be important cellular mechanism of DIC.

本实验观察内毒素在体外对兔血小板胞浆游离钙浓度[Ca~(2+)]i的作用以及用二次注射内毒素法复制DIC模型时对[Ca~(2+)]i的影响。结果表明静息血小板[Ca~(2+)]i为112±24 nM,内毒素可直接作用于血小板,使[Ca~(2+)]i呈剂量依赖性升高。内毒素致DIG时(Ca~(2+)]i可升高三倍。结果提示内毒素可能是通过升高血小板[Ca~(2+)]i而激活血小板。血小板的激活可能是导致DIC的重要发病机制之一。

The biochemical mechanism of lithium-induced hypothyroidism was studied.The incubation of intact thyroid cells with lithium for 60 min resulted in significant inhibition of both basal and agonist— stimulated cAMP accumulation.The inhibition of agonist-stimulated cAMP accumulation was dose- dependent for lithium and appeared noncompetitive.Significant inhibition was observed at 1 mmol/L lithium for forskolin-stimulated cAMP accumulation. TSH-stimulated cAMP accumulation was inhibited slightly by this concentration of lithium but not significantly.The cAMP response to all agonists, however,was significantly inhibited by 5 mmol/L lithium and higher concentrations. The reaction of TSH-induced increase in intracellular free calcium in thyroid cells was also inhibited by 5 mmol/L lithium.We came to the conclusion that lithium-induced hypothyroidism might be caused by the inhibitory effect of lithium on the principal intracellular signalling systems in thyroid cells.

The effect of endotoxin on ~(45)Ca uptake by mouse peritoneal macrophages and cytosolic free calcium in myeloma cells was studied. ~(45)Ca uptake by macrophages isolated from intravenously endotoxin-injeeted mouse was significantly increased. Macrophages from normal mouse also showed an enhanced uptake of ~(45)Ca following its in vitro exposure to endotoxin. Lanthanum chloride partially blocked ~(45)Ca uptake by both control and endotoxin-treated macrophages. The amount of lanthanum-insensitive ~(45)Ca uptake by endotoxin-treated macrophages was increased compared with that by control macrophages. Incubation of myeloma cells with endotoxin elicited a significant increase in cytosolic free calcium monitored with the fluorescent Ca~2+-indicator quin 2. The results suggest that in vivo and in vitro exposure of cells to endotoxin induces calcium transport alterations.

取自静脉注入内毒素小鼠腹腔的巨噬细胞对(45)~Ca的摄取量显著增加。取自正常小鼠腹腔的巨噬细胞在体外与内毒素温育后,(45)~Ca摄取量也有显著增加。正常巨噬细胞和经内毒素处理的巨噬细胞对(45)~Ca的摄取能被LaCl_3部分阻断,内毒素处理组不受LaCl_3影响的“Ca摄取量显著高于对照组。以钙离子荧光指示剂Quin2测定胞浆游离钙,骨髓瘤细胞经内毒素处理后,胞浆游离钙浓度显著增高。结果提示内毒素在体内和体外均能影响细胞钙转运功能,引起细胞钙平衡紊乱。
