The authors studied the changes of concentration of cytosolic free calcium within cortical neurons,activity of Ca2+-ATPase and water content of brain tissues following traumatic rat brain edema.Cerebrolysin was employed to show its effects on the calcium overload in the neurons,alteration of Ca2+-ATPase activity and brain edema.The results indicated that the neuronal Ca2+ channel opened after the brain injury.The level of cytosolic free calcium within neurons reached to 1095.42±76.15nmol/L,0.5 hours after brain injury,which was ten times the value of control.The levels of cytosolic free (Ca2+ ) i kept the highest at 6,24,48 and 72 hours after brain injury,which were 16 to 17 times the value of control.In the contrast,the activity of Ca2+-ATPase of mitochondria in the neurons reduced remarkably following the brain injury.And water contents of brain tissue increased after the injury.The en-hancment of water contents on the white matter was more evidently than that of grey matter.Cerebrolysin showed the effects on stimulating the activity of Ca2+-ATPase in the mitochondria,decreasing the levels cytosolic free (Ca2+ ) i within neurons,and giving an advantage to treatment of brain edema in this study.

本文研究大鼠脑损伤后皮质神经元突触体胞浆中游离钙浓度([Ca~(2+)]i),Ca~(2+)—ATP酶活性及脑组织水含量变化。并选用脑活素治疗,观察其对神经细胞Ca~(2+)超载及Ca~(2+)—ATP酶活性改变和脑水肿的影响。结果:脑损伤后神经元Ca~(2+)通道开放,伤后仅0.5小时,突触体胞浆中游离[Ca~(2+)]i已升高至1095.42±76.15nmol/L水平,为对照值的10倍;伤后6,24,48和72小时,[Ca~(2+)]i持续处于高水平,为对照值的16~17倍。与之相反,神经细胞线粒体Ca~(2+)—ATP酶活性显著下降。脑损伤后脑组织水含量增加,以脑白质增加较为明显。应用脑活素治疗后,线粒体Ca~(2+)—ATP酶活性有明显回升,神经元胞浆Ca~(2+)超载有所减轻,脑水肿有一定改善。

The result showed nimodipine can inhibit Ca++ influx, supress membrane phosopholipid degradation decrease the release of TXA2. The biochemistry basis of anti-calcium and anti-TXA2 treatment were discussed briefly with relative literature. Author emphasized that drugs must be used conjunctively to guard the neurons in many aspects in the treatment of cerebral ischemia.

本研究选择急性缺血性脑卒中患者38例,随机分成三组,分别给予尼莫地平、阿斯匹林以及两药伍用治疗,于治疗前、后观察血小板胞浆游离钙浓度和血浆TXB_2含量的变化,结果提示:尼莫地平能阻滞Ca~(++)内流,抑制膜磷脂降解,减少TXA_2的合成,与阿斯匹林伍用效果更佳。作者结合文献探讨了抗钙和抗TXA_2治疗的生化学基础及临床意义,提出治疗脑缺血需联合用药,在多个环节,多种途径上对神经元进行保护。

The effects of adenosine on 45Ca uptake by rat brain synaptosomal fraction (P2 fraction) stimulated by K+, Glu, and NMDA were investigated. Adenosine (10nmol/L-0. 1μmol/ L) decreased the uptake of 45Ca induced by high (60mmol/L) K+, 0. 1mmol/L NMDA, and 0. 1mmol/L Glu in a concentration-dependent manner. The maximum rates of inhibition were 41. 68 ± 7. 68% , 31. 32 ± 6. 17%, and 37. 52 ± 2. 29% respectively. The possibility of neuro-protective role of adenosine in cerebral ischemia was discussed.

用同位素标记~(15)Ca法测定大鼠突触体内游离钙的浓度,探讨了腺苷对高钾、N-甲基-D-天冬氨酸(NMDA)谷氨酸(Glu)等刺激所致的Ca~(2+)内流增加的影响。结果发现腺苷在10nmol/L~0.1μmol/L范围内对高钾、NMDA、Glu等刺激所致的Ca~(2+)内流有抑制作用,并呈剂量依赖关系,最大抑制率分别为41.68±7.68%、31.32±6.17%、37.52±2.29%。这可能是腺苷对缺血性脑损伤保护作用的机理。
