The blood flows of brain (CBF), kidney (RBF) and hind limb (LBF) were found to be respectively 130±65, 173±71 and 3.7±1.6ml/min/100g. The vascular resis:ances of brain (CVR), kidney (RVR) and hind limb (LVR) were respectively 1.0±0.4, 0.8±0.4 and 40±15 mm Hg/ml/min/100g. After an intravenous injection of the alkaloid (10—40μg/kg) isolated from Veratrum maackii, the BP, CBF and RBF all decreased. The blood flows decreased remarkably at the beginning, but slowly recovered after 5 minutes. During the hypotension, CVR and RVR showed a decline for 10—30 minutes. Intraarterial injection of 3μg/kg yielded a diminution of CBF and RBF, accompanied with an augmentation of CVR and RVR. These changes passed off in 1 minute. Hence the dilatation of cerebral and renal vessels seemed to play a certain role in the hypotensive mechanism of Veratrum maackii. Its influence on the cerebral and renal circulations were transitory and reversible. Intravenous injection of 10—20μg/kg lowered the LBF and increased the LVR. This action was demonstrated to result mainly from the muscular tetanus caused by the alkaloid.

Thirty patients with essential hypertension were randomized into 2 groups. Fifteen patients were treated with captopril 37.5-75 mg/d, po and the other 15 patients were treated with nadolol 40-80 mg/d, po, all for 4 wk. 本实验用放免方法测定雄性Sprague-Dewley大白鼠分别在结扎胆管1、2、3和4周后肾皮质与髓质组织中6-keto-PGF_(1α)及TXB_2含量来了解肾皮质与髓质中前列环素和血栓素A_2的变化。结合表明,肾皮质与髓质中6-Keto-PGF_(1α)在胆管结扎早期(1~2周)显著升高,而后期(3—4周)无继续升高。肾皮质与髓质中的TXB_2在结扎胆管后期显著升高,引起6-keto-PGF_(1α)/TXB_2比值在胆管结扎后期显著降低。提示胆管梗阻时间越长,肾组织中TXA_2合成增加越明显,导致肾脏易于出现血管痉挛,血小板聚集,肾循环功能障碍。
