Geotrichum candidum 1522 with higher lipase activity (35U/mL) was iso-lated from the mud in the Changchun grain oil processing factory. By the mutagenesis screening with ultraviolet light, LiCl, NH2OH-HCL and NTG, a mutant strain N79 with 80U/mL of lipase activity in culture broth was obtained from Geotrichum candidum 1522. Geotrichum candidum 1522 with higher lipase activity (35U/mL) was iso-lated from the mud in the Changchun grain oil processing factory. By the mutagenesis screening with ultraviolet light, LiCl, NH2OH-HCL and NTG, a mutant strain N79 with 80U/mL of lipase activity in culture broth was obtained from Geotrichum candidum 1522. The optimum medium of N79 was composed of peptone (4%), sorbitol (0.75%), olive oil (0.5%) and MgSO4·H2O (0.1%). The optimal conditions for lipase production in shaking culture: initial pH (6.0 to 6.5), temperature (26 to 30℃), aeration (30 mL of culture broth in 250 mL Erlenmeyer flask) and fermentation time (32 to 36 h). Under the optimal conditions the lipase activity was up to 105 to 115U/mL.

It shows that the protein contents of the materials can be improved through appropriate solid fermentation by analysing the fermentation conditions.The improvement of the protein contents shall be measured according to the indexes of the real protein contents and deducing the apperent increment due to the loss of materials.

通过对固体发酵条件的分析,认为合适的固体发酵可以增加发酵物的蛋白质数量,但增加量不大;在衡量固体发酵蛋白质增量时,应以真蛋白量为指标并扣除由于物料损耗而出现的表观增值,才能准确反映出真实的变化情况。
