The RC coup- ling model between the cells is established to explain the experiments.

Results The Aγ globin gene -173 T→C mutation decreased the affinity of GATA 1 to the mutant promoter fragment (-201~-158)96%( P< 0 01) and the binding of Oct 1 to the same DNA fragment 55% ( P< 0 05).The activity of the mutant promoter was 2 fold ( P< 0 05) as strong as that of the normal one in MELGM979 cells.The mutant and the normal promoters exhibited basically same activities in K562 and Hela cells. Conclusions The -173 T→C mutation decreased dramatically the binding of GATA 1,implicating that GATA 1 may act as a negative regulator of Aγ globin gene in adults.The -173 T→C mutation may enhance the activity of Aγ globin gene promoter in the adult erythroid cell environment.

目的 研究人Aγ珠蛋白基因- 173 T→C突变对反式作用因子与启动子的结合及对启动子活性的影响。方法 采用电泳迁移改变分析和荧光素酶报告基因瞬时转染分析。结果 Aγ珠蛋白基因- 173 T→C突变使GATA1 与突变启动子片段(- 201~- 158)的结合降低了96% (P< 001),并且使Oct1 与相同的DNA 片段的结合降低55% (P< 005)。在MELGM979 细胞中,突变启动子活性是正常启动子的2 倍(P< 005);在K562 和Hela 细胞中,突变和正常启动子活性基本相同。结论 Aγ启动子- 173 T→C突变导致GATA1 与突变启动子片段结合显著减少,提示在成年期该转录因子可能是Aγ珠蛋白基因的负调控因子;- 173 T→C突变可能仅在成年期红系细胞环境中才能增强Aγ珠蛋白基因启动子活性

In the present paper, a summary of the research achievements on the effects of heat shock proteins in embryo development in past ten years is given in brief and discussed.

在胚胎发育,特别是早期胚胎发育时期,基因转录活跃,蛋白质大量合成,细胞的增殖和分化非常剧烈,细胞的环境在不断变化,细胞对外界刺激十分敏感。这个时期的HSPs变化和作用表现得非常突出。 本文简要总结了近十年有关热休克蛋白在胚胎发育中作用的研究成果。根据胚胎发育对HSPs的依赖性、HSPs基因表达的发育阶段特异性和组织特异性、干扰胚胎发育中HSPs表达程序以后导致胚胎异常发育等现象推测:(1)热休克基因通过参与和/或调控体形形成、肌肉及神经分化,而在胚胎发育中具有看家基因的功能;(2)热休克蛋白作为伴侣分子,通过介导新合成的和/或可逆变性的蛋白质正确折叠、装配、转运及促进不需要的和/或不可逆变性的蛋白质降解,参与胚胎的正常发育和保护胚胎不受不良刺激的影响。这方面的深入研究,必将有助于阐明胚胎发育、细胞增殖、细胞分化和去分化、细胞转化、生物适应性等的分子机理。
