人Cu·Zn SOD是一种重要的氧自由基清除剂 ,传统工艺是从动物血中提取得到 ,受原料来源的限制。从人胎肝中提取了hCu·Zn SODcDNA构建了基因工程菌 ,同时对其表达产物的条件进行研究 ,着重探索了接种量、接种时机、pH、溶氧等因素 ,并优化了发酵条件。通过在 5L全自动发酵罐上研究重组菌BL2 1的培养条件 ,确定了最适条件为 :接种量 2 % ,溶氧水平≥ 40 % ,初始 pH6 5 ,培养过程中用 3mol/LHCl调节 pH在 7 5。在此条件下 ,发酵最高水平达到酶活95 0U/ml,比活 14 0 0U/mg。

研究了营养与环境条件对耐高温酒精活性干酵母 (TH -AADY)SOD摇瓶发酵的影响。实验结果表明 ,初糖浓度、金属离子、pH值、通气量 (装液量 )和培养时间等均对AADY摇瓶发酵的生物量和SOD含量有较大的影响。在初步优化的发酵条件下 ,细胞生物量为 39.6 g/L ,菌体SOD含量为 1 948U/g ,发酵液产酶能力为7.7万U/L。
