TMVA is a protein abstracted from Trimeresurus mucrosquamatus venom. It can induce platelet aggregation of human and many animals. The effect is realized by several ways. TMVA can induce platelet aggregation not only by increasing the release of ADP and the formation of thromboxane A2, but also by a pathway similar to that of PAF. Thus, TMVA may be used as a model for screening antiplatelet drugs. We primarily put the model to study the effects of three principles or components of Chinese medicinal herbst protopine, total saponins of panax notoginseng (SAN CHI) leaves and total saponins of panax japonicus (ZHU ZI SHEN) on rabbit platelet aggregation. The results show both protopine and total saponins of panax japonicus can inhibit rabbit platelet aggregation induced by ADP and TMVA; Total saponins of panax notoginseng leaves can only inhibit rabbit platelet aggregation induced by ADP, but not by TMVA. These are in accord with the physiological properties of TMVA and show TMVA may be used as a good model for screening antiplatelet drugs.

本文利用烙铁头蛇毒血小板聚集素(TMVA)作为筛选抗血小板药的工具,对三种中草药有效成分普罗托品,三七叶总皂甙和珠子参总皂甙进行了筛选,并与ADP模式相比较。结果发现普罗托品及珠子参总皂甙对ADP及TMVA诱导的家兔血小板聚集均有抑制作用;而三七叶总皂甙则只能抑制ADP诱导的家兔血小板聚集,不能抑制TMVA诱导的家兔血小板聚集。普罗托品抑制ADP及TMVA诱导家兔血小板聚集的IC_(50)分别为26.85±10.07μM和38.04±12.11μM。珠子参总皂甙(2mg/ml)抑制ADP及TMVA诱导的家兔血小板聚集的最大抑制率分别为51.7±4.7和50.3±4.7%。三七叶总皂甙(3mg/ml)抑制ADP诱导家兔血小板聚集的最大抑制率为44.8±4.7%。

Two new saponins, bipinnatifidusoside F_1(Ⅻ) and F_2(ⅩⅢ), were isolated along with eleven known saponins from the dried leaves of Panax japonicus var.bipinnatifidus(Seem.)Wu et Feng collected in the Range of Qinling Mountains in China. 从羽叶三七叶中分离到十三种甙类成分,经FAB-MS,~(13)CNMR谱,双照射~1HN-MR谱,~1H-~1H COSY谱及与标准品直接对照,证明十一种为已知化合物,分别为人参皂甙F_1(Ⅰ),F_2(Ⅱ),F_3(Ⅲ),R_(g2)(Ⅳ),R_a(Ⅴ),Rd(Ⅵ),R_(b1)(Ⅷ),R_(b3)(Ⅷ),24(S)-假人参甙F_(11)(Ⅸ),人参黄酮(Ⅹ)和珠子参甙F_1(Ⅺ);另外两种为新的达玛烷型皂甙,命名为羽叶三七甙F_1(Ⅻ)和F_2(ⅫⅠ),并确定其化学结构。同时修正珠子参甙F_3的结构。进一步阐明人参黄酮甙结构中的两个糖的连接方式。
