The iodonium compound (Ⅲa) could be converted to the chloride (Ⅲb), colorless needles, m.p. 211.5° (dec), bromide (Ⅲc), yellowish needles. m.p. 217.5° (dec.), iodide (Ⅲd), yellow powder, m.p. 164° (dec.) and picrate (Ⅲe), yellow needles, m.p. 183-184° (dec.) by treating its solution in formic acid with a 50% formic acid solution of sodium chloride, sodium bromide or potassium iodide or with an alcoholic solution of picric acid respectively. The structural formula (Ⅲ) of the new iodonium compounds was derived by means of potassium permanganate oxidation of the bisulfate (Ⅲa) to 2-iodo-4-nitrobenzoic acid, and by thermal decomposition of the iodide (Ⅲd) to 2,2'-diiodo-4, 4'-dinitro-diphenyl-methane (Ⅵ) (60% yield), m.p. 158-159°, the structure of which was shown by analysis and by comparison with what is known about the behavior of other iodonium iodides, e.g., 4,4'-dicarboethoxy-diphenylene iodonium iodide (Ⅷ), which decomposes on heating yielding 2,2'-diiodo-4,4'-dicarboethoxydiphenyl (Ⅸ). Compound (Ⅵ) could be reduced with tin and hydrochloric acid to a diamino compound (Ⅶ).The iodonium salt (Ⅲa) possessed the interesting property that when it was treated with dilute sodium hydroxide solution at room temperature with the addition of ethyl acetate or pyridine, a beautiful bluish-green color gradually developed in the organic layer. When it was heated with dry pyridine, the same color developed. The colored ethyl acetate solution changed to brownish yellow on acidification with dilute sulfuric acid and regained its bluish-green color when the acidified solution was made alkaline.Under similar conditions as described above, reaction between diphenylmethane and iodosyl sulfate gave a grayish substance, m.p. 210-212°, which was insoluble in common solvents. This substance was possibly a polymeric compound of formula (Ⅺ).

Reduction of three cyclic iodonium salts, 3, 6 - dinitrodibenzopyriodon-ium bisulfate (Ⅰ), 3,6 - dinitrodibenzo-p-oxo-pyriodonium bisulfate (Ⅱ)and 3, 6-dinitro-dibenzopyrroiodonium bisulfate (Ⅲ) with potassium orsodium borohydride in dimethyl formamide-methanol solution results in2-iodo-4, 4′-dinitrodiphenyl-methane, m. p. 108~9°, 2-iodo-4, 4′-dinitrodi-phenylether, m.p. 115~6°, 2-iodo-4, 4′-di-nitrobiphenyl (isolated as 2-iodo-4, 4′-diaminobiphenyl dihydrochloride, m.p. 193~4°), respectively.The conditions for reduction were examined. A new cyclic iodonium salt, 3, 6-diacetamidodibenzo-pyriodonium sulfate(Ⅵa), in.p. 208~9°, was prepared by oxidation and cyclization withpotassium persulfate in concentrated sulfuric acid from 2-iodo-4, 4′-diaceta-midodiphenylmethane, m.p. 198~200°, which in turn was obtained from2-iodo-4, 4′-dinitro-diphenylmethane by usual tin-hydrochloric acid reductionand acetylation. The diacetyl groups of the iodonium cycle (Ⅵa) werehydrolyzed with 40~50% slfuric acid, and the product was isolated as iodidesulfate (Ⅶ), m. p. 188~190°. The preliminary pharmaceutical works showedthat the 3, 6-diacetamido dibenzopyriodonium sulfate (Ⅵa) had some hypote-nsive action to dogs.

The method of estimating the sensitivity of in vitro human oesophageal carcinoma cell culture to some platinum complexes and their conjugates was used as a reasonable basis for assessment of their possible clinical sensitivity in cancer chemotherapies. It was disclosed that the cisplatin was most effective in cytostatic activity against the human oesophageal carcinoma cells (strain 109) and that the diaminodiiodoplatinum(Ⅱ), newly synthesized in our laboratory and primarily designed for attacking the thyroid malignancies, was also effective at higher concentrations which could be attainable locally owing to its inherent property of being selectively absorped by thyroid tissues. The K_2PlCl_4 and its conjugate with concanavalin A were, however, found to non-effective in this study.

观测了几种铂制剂及其结合体对我国自行建立的人食管癌细胞株(Eca 109)体外培养的抑制作用,作为它们试用于肿瘤化疗的评价基础。本实验表明我国自制顺铂对人食管癌细胞有显著的细胞毒作用。利用含碘物有被集中吸收于甲状腺组织的固有特性而设计合成的碘铂,在较高浓度时也具有明显的细胞毒作用,似可作为它试治甲状腺癌的实验室依据。至于四氯铂酸钾及其与刀豆素的结合体,在本实验条件下,并未显示任何抑制作用。
