During tissue culture of plant cells,the regeneration frequency and the quality of regenerated plants are closely related to subculture time.To determine the possibility of longterm subculture and appropriate subculture strategy,the embryogenic callus of Gossypium hirsutum L.cultivar Chuan 239 was used as material to examine the changes in color,character,growth speed and embryogenesis capability during subculture time.The morphology variation of somatic embryos and regenerated plants were also analyzed.The results showed,although the embryogenesis capability can be maintained for a long time by selective subculture on MSB medium containing KT01?mg/L,KNO319?g/L and MSB supplemented with KT01?mg/L,IAA01?mg/L alternatively,the frequency of abnormal somatic embryos and sterile regenerated plants rose with the increase of culture time.The subculture time of cotton embryogenic callus cant be more than 15 years.

植物细胞在离体培养时,再生频率和再生植株的质量与培养时间密切相关。为探讨棉花胚性愈伤组织长期继代的可行性和适宜的继代策略,以川棉239胚性愈伤组织为材料,观察其在继代过程中颜色、质地、生长速度和体胚发生能力的变化,体胚及再生植株畸形变异的发生频率。结果表明,通过在MSB+KT0 1mg/L+KNO31 9g/L和MSB+KT0 1mg/L+IAA0 1mg/L两种培养基上交替培养并结合选择性继代,川棉239体细胞胚胎发生能力虽然可保持较长时间,但随着继代时间的延长,再生能力却逐渐下降,畸形胚发生频率和再生植株不育率逐渐升高。棉花胚性愈伤组织的继代时间不宜超过1 5年。
