The gold anti-HBs conjugate reacts with HBsAg in serum and migrates up the nitrocellose strip on which anti HBs was immobilized, thus producing the detection reaction. Results 395 sera of patients and healthy people were detected for HBsAg by GICA and EIA.The corresponding rate of both tests was 99.0%. The sensitivity of GICA was 1 ng/ml by detecting HBsAg panel from the National Institute for the Control of Pharmaceutical and Biological Products. Conclusion GICA is a sensitive, specific, simple and rapid assay for detecting HBsAg.

目的建立一种简易快速、自测式胶体金免疫层析法(GICA)用于检测血清中乙型肝炎病毒表面抗原(HBsAg)。方法采用柠檬酸三钠还原法制备胶体金颗粒、标记乙型肝炎病毒表面抗体(抗HBs),制成免疫层析检测试条。血清中HBsAg与测试条上金标记抗体结合后沿着硝酸纤维素膜移动,与膜上的固相抗体结合形成肉眼可见的红色线条。结果GICA试条只与HBsAg有特异性反应,与乙型肝炎病毒核心抗原、e抗原等无交叉反应。检测中国药品生物制品检定所HBsAg标准品,灵敏度可达1ng/ml。用GICA与酶免疫法比较检测了395份不同血清标本中HBsAg,两法符合率为99.0%。结论GICA检测血清中的HBsAg特异性强、灵敏度高、简便快速,无需特殊仪器设备,有广泛应用价值。

AIM To develop a rapid, simple and self performing gold immunochromatographic assay (GICA) for the detection of anti cytotoxin associated gene A(CagA) protein of Helicobacter pylori in sera. METHODS With prepared GICA apparatus and colloidal gold which were coupled with protein A(SPA), the gold SPA conjugate reacted with human IgG in serum and migrated along the nitrocellose strip on which the CagA was immobilized. If the serum contained anti CagA IgG, there would be a red line on the strip where the CagA was immobilized. RESULTS A total of 326 sera of patients and healthy people were detected for anti CagA IgG by GICA and ELISA. The sensitivity and specificity of GICA were 95 8% and 97 3%. The corresponding rate of both tests was 96 6%. CONCLUSION GICA is a sensitive, specific, simple and rapid assay for detecting anti CagA IgG in sera.

目的 建立一种快速、简易的检测血清中抗幽门螺杆菌( Hp)细胞毒素相关蛋白 A( CagA) 抗体的胶体金免疫层析法(GICA) .方法 采用柠檬酸三钠还原法制备胶体金颗粒,标记葡萄球菌A 蛋白(SPA) ,将重组的CagA 抗原划线固定于硝酸纤维素膜上,制成免疫层析检测试条. 血清中IgG 与测试条上金标记物结合后沿着硝酸纤维素膜移动,与膜上的固相抗体结合形成肉眼可见的红色线条.结果 用GICA 与ELISA 试剂盒对比检测了326 份血清标本中抗CagA 抗体,本方法的特异性为95-8 % ,敏感性为97-3 % ,两法符合率为96-6 % .结论 GICA 检测血清中的抗CagA 抗体特异性强、灵敏度高、简便快速,无需特殊仪器设备,有广泛应用价值.

Objective:Introduce to the method of PSA McAb to label gold colloid ,to determine of PSA antigen Methods: The method using citric acid three sodium revert prepare to gold colliod,the means of immunity separate to assay.Results: To compare of the method of ELISA and GSLIA ,sensitivity ,speciality ,is not more difference than that of GSLIA,GSLIA regent places a year later in room temperature. Conclusion:It is a kind of adiagnosing PSA cancer method that GSLIA determine PSA,fast,easy,sensitive.

目的 :采用前列腺抗原 (PSA)单抗标记胶体金的方法 ,测定PSA抗原。方法 :采用柠檬酸三钠还原法制备胶体金 ,金标免疫层析方法 (GSLIA)测定。结果 :将金标方法与ELISA方法进行比较灵敏度、特异性 ,其两者无显著性差异。金标方法可在室温放置 1年以上。结论 :金标免疫层析法测定前列腺抗原 ,是一种快速、简便、灵敏的筛选前列腺癌患者的诊断方法
