    In present study, the injury induced by oxygen free radical in cultured cardiac cells was observed by adding xanthine and xanthine oxidase into the medium The effects of zinc on the injury of cells were also observed, taking the electro physiological changes and threshold concentration of BaCl_2 which induces cell stop beating as index.     In present study, the injury induced by oxygen free radical in cultured cardiac cells was observed by adding xanthine and xanthine oxidase into the medium The effects of zinc on the injury of cells were also observed, taking the electro physiological changes and threshold concentration of BaCl_2 which induces cell stop beating as index. The results showed that the electric parameters of action potential (the amplitude, overshoot, maximum diastolic potential, threshold, maximum rate of depolarization and duration of action potential ) were decreased in XOD group, compared with the control. The threshold concentration of BaCl_2 was decreased too. However, there were no significant differences between Zn group and control group except the TP value. The electric parameters of action potential was increased notab(?)y in Zn group, compared with XOD group. Furthermore, the threshold concentration of BaCl_2 in Zn group was increased too. It is suggested that Zn possessed the protective effects on cultured cardiac cells from the injury induced by oxygen free radical.

    本文通过向培养基中加入黄嘌呤和黄(?)呤氧化酶系统成分造成培养的心肌细胞受氧自由基伤害,以电生理的改变和BaCL_2引起心肌细胞停搏的闭浓度为指标,观察了微量元素Zn的抗氧化损伤作用。结果示:XOD组与对照组比较,动作电位APA、OS、MDP、TP、Vmax及APD_(50)各电参数明显减小。BaCl_2引起心肌细胞停搏的阈浓度亦减低。而加Zn组与对照组比较,以上结果除TP值偏低P<0.05外,其它参数则无显著性改变。加Zn组与XOD组相比,动作电位各电参数明显增大,BaCl_2引起心肌细胞停搏的阈浓度亦增高。提示Zn对培养的心肌细胞氧自由基所致的损伤具有保护作用。

    When cultured cardiac cells of rats were exposed to X-XOD containing medium, the absolute values of electronic parameters were decreased the percentange of beating clusters reduced and the microscopic structure destroyed. PQS significantly converted all the indices, which shows that PQS can protect the cells from oxidative damage.

    黄嘌呤-黄嘌呤氧化酶可引起大鼠培养心肌细胞电参数变小、心肌细胞起搏比例减少及心肌细胞超微结构破坏。西洋参茎叶皂甙可使上述指标明显改善,说明其有抗氧化损伤作用

    Using the methods of observation under electron microscopy and electron spin reso- nance spectroscopy (ESR). we observed the damage action of X -XOD and antioxidative damage action of panaxadiol saponin monomer Rb1 on the cultured myocardiocytes in neonatal Wistar rat. The results indicated that X - XOD destroyed the myofilaments and mitochondria and increased the amount of free radicals. Rb1 30μg/ml added to the medium could apparently antagonize all of the variations induced by X-XOD.

    采用电镜观察和电子自旅共振(ESR测定)的方法,我们观察了黄漂呤—黄嘌呤氧化酶(X—XOD)对培养新生Wistar大鼠的氧化损伤作用以及人参二醇组皂甙单体Rb_1抗氧化损伤作用。结果表明,X-XOD增加自由基数量,破坏肌丝和线粒体等作用。加入培养基中Rb_130μg/ml能明显对抗X-XOD引起的各种变化。
