To evaluate the forensic applicability of STR loci D19S253 and D8S1179,studies were performed using PCR and PAG vertical electrophoresis. The experiments included: mixed-stains analysis; typing of DNA in blood samples deposited on various substrates; determination of the sensitivity of two loci; determination of cross-reactivity of DNA from species other than human and DNA typing of different tissues from the same individual. The results demonstrated that both D19S253 and D8S1179 loci are of high species specificity...

To evaluate the forensic applicability of STR loci D19S253 and D8S1179,studies were performed using PCR and PAG vertical electrophoresis. The experiments included: mixed-stains analysis; typing of DNA in blood samples deposited on various substrates; determination of the sensitivity of two loci; determination of cross-reactivity of DNA from species other than human and DNA typing of different tissues from the same individual. The results demonstrated that both D19S253 and D8S1179 loci are of high species specificity with the exception of rabbit and their sensitivity of detection is 0. 25ng and 0. 5ng respectively. These two STR typing method are reliable, robust and efficient, and provide two useful tools in forensic case work.

为评估STR位点D19S253和D8S1179的法医学应用价值,应用PCR和PAG垂直电泳技术对两位点的种属特异性,检测灵敏度,以及同一个体不同组织分型的同一性及不同基质和不同保存时间的斑痕分型等与法医应用有关的问题进行了研究,D19S253和D8S1179位点的检测灵敏度分别为0.25ng及0.5ng,同时两位点具有较高的种属特异性,同一性及较好重复性,且能够复合扩增,表明D19S253和D8S1179是法医学检案中较实用的两个STR标记。

Laser was applied to revealing the scope of semen stains and mixed stains. Morphological examination and an analysis of amplified fragment length polymorphism of D 1S 80 locus are proved to be helpful in promptly solving a case.

应用激光显现法医生物物证中常见棘手的微量精斑以及混合斑斑痕范围, 通过形态学检测确证.用多聚酶链反应, 小型聚丙烯酰胺凝胶电泳和银染方法, 检测其 D N A、 D1 S80 位点扩增片段长度多态性, 与相关物证作比较, 以达到快速破案的目的. 探讨了结合激光检测微量精斑以及混合斑的法医应用, 并应用于实际案例的检验.
