    Morphology of cells of long-term cultured pulmonary squamous carcinoma cell line (LTEP-78) and cancer cells taken from sputa of fifty lung cancer patients was studied in view of clinical cytology. The results showed that the long-term cultured cell line still have had the characteristics of squamous cell carcinoma, such as round or squamous cell outline, definite cellular border, evenness in density of cytoplasm, centrally placed nuclei and so on.     Morphology of cells of long-term cultured pulmonary squamous carcinoma cell line (LTEP-78) and cancer cells taken from sputa of fifty lung cancer patients was studied in view of clinical cytology. The results showed that the long-term cultured cell line still have had the characteristics of squamous cell carcinoma, such as round or squamous cell outline, definite cellular border, evenness in density of cytoplasm, centrally placed nuclei and so on. On the other hand, it appeared that they have undergone some changes which resembled to adenocarcinoma cells, such as malignent signet-ring cell. We concluded, therefore, that the long-term cultured cancer cells could still keep the characteristics as cancer cells taken freshly from lung cancer patients. The differences from the fresh lung cancer cells found in cultured cells could be explained by the effects of in vitro circumstance which is quite different from that of the human body.

    本文报告了从临床细胞学观点对长期体外培养的人肺癌细胞进行形态学研究的结果。发现虽然经长达一年半的体外培养,人肺鳞癌细胞系仍保留有鳞癌细胞临床诊断的标准特点。大多数细胞仍为圆形或多边形;边界清楚;核位于细胞中央;胞浆匀实;并出现典型的鳞癌细胞,如圆形癌细胞、蝌蚪形癌细胞及多核癌细胞等。在培养细胞中也出现细胞分化状态的改变,如少数细胞保存着低分化鳞癌细胞的状态,甚至呈未分化的恶性裸核,而大多数细胞转化为中分化的鳞癌细胞。值得注意的是,在少数培养细胞中出现腺癌细胞样的改变,如癌细胞的胞浆内出现单个或多个大空泡,并将核推到一边。有的细胞呈典型的恶性“印戒细胞”。因此,我们认为体外培养的人癌细胞仍可反映出人体癌细胞的临床诊断特点,所出现的差异可能是体外环境选择而使细胞改变的结果。

    作者采用 ̄51Cr4h释放试验和裸鼠移植瘤模型,从颈淋巴清除术后标本中制取肿瘤引流区淋巴结细胞(DNL),经体外激活后与自身LAK细胞比较对人舌鳞癌细胞系Tca8113细胞体外、体内的杀伤活性,结果显示:E:T为25:1时,DNL组体外杀伤率为65%,LAK组为28%,按Winn氏中和法分别将DNL,LAK细胞同Tca接种于裸鼠皮下,结果DNL组抑瘤率为74%,LAK组为42%,经统计学分析两组间存在显著性差异(P<0.05),提示DNL细胞杀瘤活性明显强于LAK细胞,且来源容易,与传统的LAK细胞比较在癌过继免疫治疗中有较高的实用价值。

Tke injured target cell had a highly condensedcytoplasm and chromatin and formation of cytoplasmicblebs, which were consistent with an apoptotic celldeath rather than with a lytic or necrotic cell death. Themechanism was exocytosis of granules containing a pore-forming protein. Holes formed in tlie target cell’s mem-brane may allow a lethal substance to enter and to in-duce its death via an apoptotic mechanism.

    以人基因重组白细胞介素2(rIl-2)激活的人口腔癌引流区淋巴结细胞(DNL),具有杀伤人舌鳞癌细胞系Tca8113细胞的作用。在光镜下观察到,当二者共同培养12天后,形成似玫瑰花环状形态,靶细胞出现水疱变性。透射电镜下观察到,DNL细胞形态呈多样,核远离与靶细胞接触的一侧;其胞浆内出现较多的分泌颗粒和线粒体,且朝向细胞伸出伪足和突起与靶细胞接触的一侧。靶细胞受攻击后表现为“凋零状坏死”(apoptosis)。本研究结果提示,DNL细胞的杀伤机理之一,可能通过直接接触释放特定物质,使肿瘤细胞“穿孔”,发生通透性改变,最终导致与一般病理状态下细胞死亡有所不同的凋零状坏死。
