PURPOSE To provide experimental animal modal of impotence with diabetes mellitus (DM) for clinical and experimental researches. METHODS Sprague-Dawley rats were injected intraperitoneally with streptozotocin (STZ)to make experimental models of DM. PURPOSE To provide experimental animal modal of impotence with diabetes mellitus (DM) for clinical and experimental researches. METHODS Sprague-Dawley rats were injected intraperitoneally with streptozotocin (STZ)to make experimental models of DM. DM was diagnosed by clinical characteristies, urine glucose,blood glucose, and HbAlc.Penile erections and yawns of the DM, control(NDM)and STZ control group(STZ) after apomorphine(APO) injection were observed at the sixth, eighth and twelfth weeks starting from the fourth day of injecting STZ. RESULTS Compared with NDM or STZ group,the ratio of penile erections and the number of penile erections and yawns of the DM group decreased significantly, no significant difference between the NDM and STZ groups was observed decreased. The ratio of penile erections of the DM group decreased significantly with the increasing of the time course. CONCLUSIONS APO injection can be used to select animal model of impotence in the experimental DM model for clinical and experimented researches.

目的为临床及科研提供糖尿病(DM)性阳萎动物模型。方法应用sprague-dauley大鼠腹腔内注射链脲佐菌素(STZ)制造DM动物模型。根据临床表现,尿糖、血糖及糖化血红蛋白来综合诊断DM。注射脱水吗啡(APO)后观察6周(Ⅰ组)、8周(Ⅱ组)、12周(Ⅲ组)时,DM组、空白对照组(NDM)及STZ药物对照组(STZ)阴茎勃起及打呵欠的情况。结果3组中DM组与NDM或STZ组比,阴茎勃起率、阴茎勃起次数及打呵欠次数明显降低(P<0.001,P<0.01及P<0.01);而NDM与STZ组相比无显著性差异(P>0.05)。3组DM组阴茎勃起率两两相比有显著性差异(P<0.05)。即随DM病程延长,阴茎勃起率明显下降。结果提示DM严重影响阴茎勃起功能,且与其病程明显相关。在DM动物模型的基础上注射APO可为临床科研筛选DM性阳萎动物模型

To research the mechanisms of erectile dysfunction(ED) with diabetes mellitus (DM),streptozotocin(STZ) was injected intraperitoneally into Sprague Dewley rats to make experimental models of DM.Injected with apomorphine,penile erections were observed and noted on the 6th week,8th week and 12th week from the 4th day of injecting STZ and experimental models of ED with DM were selected.The concentration of LH,FSH and testosterone were examined and the microstructrure of testes were observed.The concentration of testosterone decreased significantly and the sigificant pathological changes much related to the duration of DM were observed in ED rats with DM.The concentration of LH did not change in the early duration of DM,but decreased significantly in the late duration of DM.No changes of the concentration of FSH were observed.The results indicated the function of penile erection and the synthesis of testosterone were affected seriously by DM,and the decreasing of the concentration of testosterone may be one of the most important mechanisms.

SD大鼠注射链脲佐菌素制造糖尿病 (DM)动物模型后 ,注射阿朴吗啡观察不同时期大鼠阴茎勃起情况 ,筛选DM性阴茎勃起功能障碍 (ED)大鼠模型 ,测定其血清LH、FSH及睾酮的浓度 ,并观察睾丸组织的显微结构 ,研究DM性ED的发病机理。结果DM性ED大鼠血清睾酮浓度显著降低 ,睾丸组织显微结构发生明显病理性改变 ,且上述变化与DM病程密切相关 ;LH在DM早期无改变、晚期明显降低 ;FSH无明显改变。提示DM严重影响阴茎勃起功能及雄激素的合成分泌 ,雄激素降低可能是其主要发病机理之一

Objectives: To investigate the effect of diabetes mellitus on nNOS containing nerve fibers in rats corpus cavernosum. Methods: Healthy male Sprague Deuley rats ( n =34) were divided into two groups randomly, the sixth ( n =17) and eighth groups ( n =17). Eleven of severteen rats wree intraperitoneal injection with streptozotocin (STZ)to induce experimented model of diabetes meditus in each group. The other were injected with citrate buffer as control. Injected with apmorphine at sixth and eight week,penile erection in diabetic and control groups were observed and noted. The expression of nNOS positive containing nerver fibers in corpous cavernosum were examined with streptavidin peroxidase immunohistochemistry (SP method). Results: The erection rate of penile diabetic mellitus decreased compared to that in controls ( P <0.01). The erection rate of two diabetic groups were 37.5% and 14.3%,respectively, with significant difference ( P <0.05).The controls were all 100%. The sigificiant changes of nNOS containing never fibers were observed in the two diabetic groups. The numbers of nNOS containing nerver fibers in the corpus cavernosum in the two diabetic groups were (37.00±6.76) and (28.00±5.29),respectively, with significant difference compared to each other ( P <0.05). In the two controls, the fibers were (83.00±3.22) and (81.00±3.61),respectively. The fibers in diabetic groups were less than those in controls( P < 0.01 ). Conclusions: The erectile function of rats and the number of nNOS positive containing nerve fibers is affected seriously by diabetes mellitus and it is positively correlated to the duration of diabetes mellitus.The decreasing of nNOS positive containing nerve fibers may be one of the mechanisms of diabetic erectile dysfunction.
