Afterwards, the blood samples were collected at regular intervals. The plasma progesterone concentration in this group was lower than that in the control group but higher than that injected with l5-Me-PGF22a alone. It showed that the luteo-lytic effect of l5-Me-PGF2a was reduced after hysterectomy.The ovarian tissue of the seventh day pregnant rats was incubated with uterine tissue and/or 15-Me-PGF2a. The ovarian progesterone concentration in the uterine tissue group or 15-Me-PGF2a group was not different from that in the control; while in the group of uterine tissue added together with 15-Me-PGF2a, the progesterone concentration was significantly decreased when compared with the control. It showed that the progesterone biosynthesis by ovarian tissue incubated in vitro would be inhibited by the existence of 15-Me-PGF2a together with uterine tissue.15-Me-PGFa, or PGF2a did not affect the ovarian progesterone concentration when added into the culture. It implied that the two drugs had no direct effect on progesterone biosynthesis by pregnant rat ovarian tissue.Human chorionic gonadotrophin (HOG) obviously increased the ovarian progesterone concentration when added into the oulure alone. However, when 16-Me-PGF2a, was added together with HOG, the ovarian progesterone concentration dropped to the level similar to that of the control. Thus 15-Me-PGF2a was shown to block the action of HOG at the level of ovary.

    It is known that human chorionic gonadotropin (hCG) elevates cAMP concentration in the ovaries of immature female rats and increases the weight of seminal vesicle of immature male rats.     It is known that human chorionic gonadotropin (hCG) elevates cAMP concentration in the ovaries of immature female rats and increases the weight of seminal vesicle of immature male rats. In the present paper, these two effects of hCG were used as indexes to examine the anti-hCG effect of ethane-1,2-dimethyl-sulfonate (EDS) and its derivatiVes. Results indicate that:(1) EDS (75~100 mg/kg), chloroethyl methane sulfonate (75~100 mg/kg) or bromoethyl methane sulfonate (25~50mg/kg) exhibited significant antagonistic action to hCG in both test models, while glycerol trimethane sulfonate (75~120 mg/kg) or propylene-1,2-dimethane sulfonate did not show any anti-hCG effect.(2) Ethyl-methane sulfonate (120 mg/kg) or β-phenyl-ethyl-methane sulfonate (120mg/kg) revealed suppressive effect on hCG in seminal vesicle weight test. However, at the same dosage, these two compounds did not show apparent anti-hCG action in cAMP concentration test.(3) n-Propyl-methane sulfonate (75~120 mg/kg)or isopropenyl-methiane sulfonate (120mg/kg) exhibited anti-hCG effect in seminal vesicle weight test.According to the data presented, the possible loci of action and the structureactivity relationship of EDS and its derivatives were discussed.

    用人绒毛膜促性腺激素(hCG)升高幼大鼠卵巢cAMP含量及增加幼大鼠储精囊重量的方法,试验了乙二醇二甲磺酸酯及其8种衍生物的抗hCG作用。结果表明:乙二醇二甲磺酸酯、甲磺酸氯乙酯及甲磺酸溴乙酯都有明显抗hCG作用;甘油三甲磺酸酯、丙二醇-1,2-二甲磺酸酯则无此作用;甲磺酸乙酯及甲磺酸-β-苯乙酯有抗hCG的储精囊增重作用,但对卵巢cAMP浓度无明显影响;甲磺酸正丙酯及甲磺酸异戊酯在储精囊增重试验中,表现了抗hCG作用。根据以上实验资料,初步分析了乙二醇二甲磺酸酯类抗hCG作用部位及构效关系。

    In the present study, 7α-methyl-10β, 17β-diacetoxy-A4-estren-3-one were shown to be able to terminate pregnancy following subcutaneous administration on the 7 th and 8 th days of pregnancy in rats, and on the 4 th and 5 th days in mice. After administration of effective dose of 7α-methyl-10β, 17β-diacetoxy-△~4-estren3-one to early pregnant rats, the peripheral plasma progesterone level was decreased markedly. At the concentration of 10 μg/ml, 7α-methyl-10β, 17β-diacetoxy-△~4-stren-3-one showed significant inhibiting action on progesterone biosynthesis in isolated pregnant rat ovary. Relative binding affinity (RBA) of 7a-methyl-lOft, 17β-diacetoxy-△~4-estren-3-one for rabbit uterine estrogen receptor was found to be 10.8 and that of 7β-methyl-10β, 17β-diacetoxy-△~4-estren-3-one to be 1.5. However, 7△-and 7β-methyl-10β, 17β-diacetoxy-△~4-estren-3-one were of poor affinity for progesterone binding sites. Result obtained from the uterine growth test in immature mice showed that the estrogen and antiestrogen activity of 7β-methyl-10β, 17β-diacetoxy-△~4-estren-3-one was weaker than that of 7α-methyl-10β, 17β-diacetoxy△~4-estren-3-one.

    7α-和7β-甲基-10β,17β-二乙酰氧基-△~4-雌甾烯-3酮(简称7α-和7β-甲-乙氧雌酮)对小鼠抗早孕ED_(50)分别为1.6和5.5 mg/kg。7α-甲-乙氧雌酮在大鼠也有抗早孕作用并使血浆孕酮浓度降低,应用10 μg/ml浓度能抑制离体妊娠大鼠卵巢孕酮合成。7α-和7β-甲-乙氧雌酮与兔子宫胞浆雌二醇受体的相对结合亲和力(RBA)分别为10.8和1.5,与孕酮受体的RBA均<1.7α-和7β-甲-乙氧雌酮都有较弱的雌激素和抗雌激素活性。
