用第8因子相关抗原(FⅧRAg)、纤维连接蛋白(FN)、Ⅳ型胶原、HLA-DR抗原和巨细胞病毒抗原(CMV)5种标记抗体,对12例新疆Kaposi肉瘤(KS)瘤组织进行了免疫电镜观察。结果表明,KS组织内FⅧRAg、FN及Ⅳ型胶原均有阳性表达,特别是FⅧRAg主要见于明细胞内,暗细胞极少,FN在明、暗两种瘤细胞及间质中均可见到,Ⅳ型胶原主要见于少数基膜上,这一结果进一步证明KS虽是一血管肿瘤,但可能不是单一的内皮起源,而是起源于血管内皮和周细胞的观点。同时在12例KS组织内均见有CMV和HLA-DR抗原的胶体金颗粒沉积,而作为对照的8例白癜风皮损除在真皮浅层见有少数树突状HLA-DR阳性细胞外,CMV抗原均为阴性,它提示CMV感染可能在KS的发生过程中起着重要作用

Objective To get more insight on the molecular mechanisms involved in psoralen induced melanogenesis during repigmentation of the hypomelanotic lesions of vitiligo vulgaris under PUVA treatment. Methods The effects of various concentrations of 8 methoxypsoralen (8 MOP)(10~500μmol/L) upon cellular morphologic changes, tyrosinase activity, and melanin contents of cultured B16F10 murine melanoma cells were examined in vitro. Results 8 MOP caused a striking change in cellular morphology with many elongate cell dendrites and deposits of brown granular materials in cytoplasm, these effects were observed clearly at final concentrations between 50 and 100μmol/L of 8 MOP after treating for 48 hours. In a range of final concentrations(10~500μmol/L), there were a dose dependent stimulation of tyrosinase activity and melanin contents with a 2~3 fold increase over untreated cells ( P< 0.01 ), the maximum action was obtained at 50 and 100μmol/L. It was also found that there was a dose related inhibition of cell growth with a 60%reduction of growth at a final concentration of 100μmol/L and no growth of cells at the concentration of 500μmol/L. Furthermore, isobutyl methylxanthine(IBMX), a protein kinase A (PKA) activator, potentiated the 8 MOP upregulation of tyrosinase activity and melanin contents, however, 12 O tetradecanoyl phorbol 13 acetate (TPA),a potent activator of PKC, inhibited the upregulative effect of 8 MOP when treated for a long duration. Conclusion These results demonstrate that in murine melanoma cells 8 MOP could induce melanogenesis by increasing tyrosinase activity and melanin contents. At least, PKA and PKC-dependent signaling pathways appear to play an important role in the melanogenic effect of 8 MOP.

Objective To explore the regulatory mechanism of tyrosinase activity by cAMP/PKA dependent signaling pathway. Methods The regulatory effects on tyrosinase mRNA level were assessed by forskolin, a specific activator of adenylate cyclase in cultured B16F10 murine melanoma cells by using RT-PCR technique. Results It was shown that forskolin could significantly enhance tyrosinase activity, melanin content, and tyrosinase mRNA level. Tyrosinase activity and melanin content were as much as seven to ten-fold higher in cells exposed simultaneously to forskolin and 8-MOP than those in untreated cells. The levels of tyrosinase mRNA were up-regulated by 26.06% with 8-MOP, 6.58% with forskolin, and 56.82% with 8-MOP plus forskolin, respectively. Conclusion Forskolin is shown to enhance 8-MOP-induced tyrosinase activity, probably by way of up-regulation of tyrosinase mRNA expression and tyrosinase synthesis. Further study is needed to understand the therapeutic significance of forskolin for vitiligo.

目的 观察蛋白激酶 A信号途径对 8-甲氧补骨脂素 (8-MOP)诱导的鼠黑素瘤细胞黑素生成及酪氨酸酶基因表达水平的影响。方法 用半定量逆转录聚合酶链反应技术检测腺苷酸环化酶特异性激活剂毛喉素对 8-MOP诱导的鼠黑素瘤细胞酪氨酸酶活性、黑素含量以及酪氨酸酶基因表达水平的影响。结果 毛喉素与 8-MOP联合处理组所测的酪氨酸酶活性和黑素含量与空白对照组相比提高近 7或 10倍。毛喉素与 8-MOP单独处理组较空白对照组酪氨酸酶 mRNA表达水平上调百分率分别为 26.06%和 6.58%,而联合处理组上调百分率为 56.82%。结论 毛喉素在体外对 8-MOP诱导的黑素生成呈明显增强作用,这一作用与毛喉素在转录水平上调酪氨酸酶 mRNA表达促进酶蛋白合成有关。毛喉素作为治疗白癜风的一种信号转导药物,其临床应用价值有待今后进一步研究。
