We have modified and improved the method of immune adherence hemagglutination (IAHA) test and used it to detect virus antigen and antibody of epidemic hemorrhagic fever (EHF). We compared the IAHA titers of different strains infected in Vero-E_6 cells The IAHA titer of A_9 strain showed to be the highest (1: 64) among the three strains. 9 EHF patients' paired sera, 3 con valecant sera and 6 immunized rabbits' sera were detected with IAHA and compared with IFA. The positive rates detected by the two tests were just the same. But their antibody titers and fold increase with IAHA were 2—6 times higher than those with IFA. These experimental results indicated that IAHA could be used as an alternative of IFA to detect EHF virus antigen or antibody in the samples from patients or infected animals.

本文对免疫粘附血凝(IAHA)技术进行了改进并用于检测流行性出血热病毒搞原和血清效价。比较了不同毒株在Vero-E_6细胞内抗原滴度,表明A_9株的滴度最高达1:64,以IAHA法对出血热病人9份血清,3份恢复期血清和家兔免疫血清6份进行了效价测定。并同时用免疫荧光(IFA)法进行比较,结果抗体阳性率二种方法一致,但前者的抗体滴度和增长倍数较后者高2-64倍,以上结果表明IAHA法可代替IFA法用于出血热病人或感染动物的抗体检测。

Serotypic classification of HFRS viruses isolated from rodents and patients were investigated using plaque reduction neutralization test ( PRNT ) . Sera of rabbits inoculated with different HFRS viruses were tested against reference type-1 Hantaan virus and type-2 Seoul virus. The neutralizing titers of antisera of Apod emus isolates against type-1 Hantaan virus were 16-fold higher than those for type-2 Seoul virus, while the antisera of Rattus isolates had titers to type-2 viruses 2-16-fold higher than those for type-1 viruses. So different isolates could be classified into serotype 1 (consistent with Hantaan virus) and serotype 2 (consistent with Seoul virus) .We also found two isolates, one from Rattus, another from patient, which could be classified into serotype 1 by PRNT, which is inconsistent with data previously published.

应用空斑减少中和试验对分离自不同动物宿主和病人血液的流行性出血热病毒进行病毒抗原性分型,以不同株的家兔免疫血清对国际上血清1型和2型参考毒株进行试验。结果表明,3株来自黑线姬鼠的毒株,其免疫血清对血清1型(76-118株)的效价较血清 2型(UR株)高≥16倍,而6株来自家鼠和大白鼠的毒株中,有5株对2型的血清效价高于1型2~16倍。提示来自黑线姬鼠的毒株符合国际上的血清1型,来自褐家鼠的毒株基本与国际上的血清2型一致,但个别毒株例外。以上结果表明,空斑减少中和试验特异性高,可用于流行性出血热病毒的准确血清学分型。 本文还发现,J10株(分离自锦州市褐家鼠)和HB55诛(分离自河南出血热病人)的血清型与过去文献报道的不符。经反复用不同免疫血清和型特异性单克隆抗体做空斑减少中和试验,均表明这两株病毒的分型与血清1型相同。以上结果为我国首次应用空斑减少试验对我国出血热不同毒株进行准确分型的报道。

The results indicated that except one strain(K_(24)), all the 13 isolates could be classified into either serotype 1 or serotype 2 according to the differences(≥4 fold) of the neutralizing titers between the sera reacted against reference type 1 and type 2 viruses, However the isolate K_(24) from R novegicus could be classified as type 2 by using known type 1 and type 2 virus antisera for neutralization with the K_(24) virus. In this study the K_(24) strain was shown to be an antigenically broad HFRS virus as K_(24) antisera reacted with 6 type 1 and 4stype 2 strains to similar neutralizing titers(≤2-fold difference).

本工作进一步对分离自不同来源的14株出血热病毒用空斑减少中和试验(PRNT)方法进行了分型。以不同株的家兔免疫血清对国际上血清Ⅰ型和Ⅱ型参考毒株进行试验,除分离自褐家鼠的K_(24)-株外,其余13株均可定为血清Ⅰ型或血清Ⅱ型用已知的Ⅰ型和Ⅱ型出血热高价免疫血清与K_(24)株进行中和试验,根据两型免疫血清的中和效价则可将K_(24)株定为血清Ⅱ型,但是K_(24)株免疫血清对6株Ⅰ型病毒和4株Ⅱ型病毒的中和效价几乎相同(相差≤2倍),表明K_(24)株是一株具有广谱中和抗原的出血热病毒。
