A protective effect of Radix Angelicae sinensis (RAS) on the liver in rat with chronic CC14-induced liver injury was confirmed. In order to elucidate its mechanism, liver ultra-structure of the above animals was demonstrated by transmission electron microscopy. The results showed that in group CC14 there were severe hepatocyte degeneration and necrosis, and a huge amount of collagen fibers remained within the necrotic focus. A protective effect of Radix Angelicae sinensis (RAS) on the liver in rat with chronic CC14-induced liver injury was confirmed. In order to elucidate its mechanism, liver ultra-structure of the above animals was demonstrated by transmission electron microscopy. The results showed that in group CC14 there were severe hepatocyte degeneration and necrosis, and a huge amount of collagen fibers remained within the necrotic focus. The survival of hep-atocytes were small in size and had little change of hepatocellular apparatus. In group CC14 plus RAS, necrotic hepatocytes were seldom seen and mitochondria and endoplasmic reticuli significantly proliferated. The interhepatocellular space of great width was always observed in group CC14, in which filled with a great number of swollen microvilli, necrotic apparatus and collagen fibers. The cavity of hepatic sinusoid stenosed and obliterated, the capillarization of liver sinusoid and the collagenization of Disse space were often observed.But, there were no these changes in group CC14 plus RAS. The present study suggests that RAS has a remarkably protective effect on the liver ultrastructure in the rat with CC14 -induced liver in- jury, and its mechanism may be improvement of hepatic microcirculation.

采用大鼠慢性CCl_4肝损伤模型观察当归对其肝脏超微结构的影响。结果表明,CCI_4组大鼠肝脏有大量肝细胞变性坏死,坏死灶中可见大量胶原纤维,残存肝细胞体积缩小,细胞器变化轻微,CCl_4加当归组肝细胞坏死偶见,肝细胞内线粒体、内质网明显增生。CCl_4组肝细胞间隙极度增宽,其内为肿胀的微绒毛、坏死物质和胶原纤维,肝窦腔明显狭窄或角塞,有明显Disse间隙胶原化及肝窦毛细血管化:CCl_4加当归组也有肝细胞间隙增宽,其内主要为增多、延长的微绒毛,坏死物少见,没有胶原沉积,肝窦通畅。研究证实,当归对CCl_4所致的慢性肝损伤的肝脏超微结构有明显保护作用,其作用机理可能主要与改善肝脏微循环有关。

Hepatocyte degeneration and inflammatory reaction were distinctivelyinhibietd,and deposition of type Ⅰ、Ⅲ、Ⅳ、Ⅴ collagen and desmin was significantly decreasedwhile sinusoid microcirculation was restored after LCSEM treatment. This indicates LCSEM hasdissolvent and absorbent efficacy to collagenous fiber which has deposited in the liver. Therefore,hepatic fibrosis might be reversed. The antifibrosis of LCSEM is more effective than colchicine(P <0.01 ). Additionally, CCl4-induced fibrosis of rats could somewhat restore. It suggestedthat LCSEM could significantly reduce CCl4-induced fibrosis of rats.

应用清肝化瘀口服液治疗小剂量CCl_4诱导的肝纤维化大鼠,并与正常对照组、CCl_4模型组、自然恢复组、秋水仙碱治疗组对照。对各组肝脏Ⅰ、Ⅲ、Ⅳ、Ⅴ型胶原及结蛋白进行免疫组化染色和计算机图像分析,并用电镜观察肝脏超微结构的变化。结果发现,清肝化瘀口服液可抑制肝细胞变性坏死,减轻炎症反应,改善微循环,抑制Ⅰ、Ⅲ、Ⅳ、Ⅴ型胶原及结蛋白在肝内的沉积,并可使已形成的胶原重新溶解和吸收,使肝纤维化逆转,效果优于秋水仙碱(P<0.01);同时,CCl_4诱导的大鼠肝纤维化可有一定程度的自然恢复。表明清肝化瘀口服液对大鼠肝纤维化有显著的治疗作用。

The immunohepatic fibrosis model was induced by intraperitoneal injection of porcine serum, treated with the TCM Decoction whose function was removing heat and dampness, nourishing qi and spleen, tonifing blood and motivating blood circulation. 用猪血清腹腔注射法复制大鼠免疫性肝纤维化模型,用以清热利湿、补气健脾、养血活血为主要治法的中药2号方进行预防和治疗,用光镜观察肝脏HE、Malory染色的变化,用电镜观察肝脏超微结构的变化,并检测了血清生物化学的变化。结果证明中药2号方有明显的改善肝功能和抗肝纤维化的作用
