AIM To identify whether the passage animal cell lines had neoplastigenesis in the nude mice. METHODS MDCK,Vero,F 81 ,BHK 21 and MA 104 cell lines were cultured respectively and harvested during logarithm growth, and then were counted and resuspended sterily before they were inoculated against nude mice. After a period of growth in nudemice, these cells were evaluated for neoplastigenesis by pathoanotomy and pathomorphology. RESULTS All of the nude mice had neoplasms after inoculation by the vero,F 81 , MA 104 passaging cells. The neoplasms were inflammatory tissue by pathological morphology, and their oncogene ratio was zero. While MDCK's oncogene ratio was zero inside the 30 passage, in the 30~60 passage cells, it became 10~40% and BHK's oncogene ratio was 100%. CONCLUSION F 81 ,vero,and MA 104 belong to non oncogenous cells. MDCK is a type of low oncogenous passage cell, but after being freezed melted and cleavaged for 3 times or disposed with supersonic wave for 5 min, no oncogenesis can be observed, However,BHK 21 can produce malignant tumor in nude mice after inoculation subcutaneously 9~21 days and has a stable repeatability.

目的 观察 5种动物传代细胞系对裸鼠是否具有致瘤性 .方法 犬肾细胞系 (MDCK)、非洲绿猴肾传代细胞系(Vero)、猫肾细胞系 (F81 )、地鼠肾传代细胞系 (BHK2 1 )和恒河猴肾细胞系 (MA- 10 4)分别培养后 ,选择对数生长期的细胞进行消化、离心、计数后 ,制备细胞悬液 ,接种裸鼠 ,定期观察 ,通过病理组织形态学观察判定结节是否具有肿瘤的特性 .结果  Vero、F81 、MA- 10 4接种裸鼠后全部有肿块形成 ,经病理形态学检查 ,均为炎性组织 ,其致瘤率为零 .MDCK在 30代以内 ,致瘤率为零 ,第 30~ 6 6代致瘤率为 10 %~ 40 % ,BHK2 1 致瘤率为 10 0 % .结论  F81 ,Vero和 MA- 10 4属非致瘤性细胞系 ,MDCK属低致瘤性细胞系 ,但经冻融或超声波裂解后对裸鼠无致瘤性 ,BHK2 1 属致瘤性细胞系 ,其正常细胞经裂解后仍具有致瘤性 ,而且具有稳定的重复性

A canine adenovirus (CAV) strain was isolated from a diarrhea dog with MDCK cell, it was demonstrated that it is a type 2 virulent strain, by systematic identification and was named SY. A canine adenovirus (CAV) strain was isolated from a diarrhea dog with MDCK cell, it was demonstrated that it is a type 2 virulent strain, by systematic identification and was named SY. Since it can induce high SN antibody titer in dogs against CAV, it may be a potential vaccine strain candidate. The virulent SY strain was attenuated by serial passages in MDCK cell line. The pathogenicity of SY strain was tested by experimental infection in susceptible dogs with the virus on 15th, 13th, 45th and 60th passage lever, respectively. The result showed that the pathogenicity of SY strain decreased gradually with the increasing of passages and lost pathogenicity to dogs of the 60th passage.

作者对分离和系统鉴定的一株犬传染性喉气管炎病毒 (犬 2型腺病毒 ) SY(沈阳 )株 ,用犬肾传代细胞 MDCK进行了致弱驯化 ,每驯化 1 5代做一次初步的犬的安全性试验 ,用大剂量不同代次病毒对易感犬进行人工感染试验 ,每天观察犬的临床症状及白细胞总数 ,并于接毒后的第 5 d安乐死 ,作病理解剖学和病理组织学检查。易感犬的感染试验结果表明 ,SY株在犬肾细胞上传 1 5代时对犬的致病力已降低 ,3 0代时对犬已不引起发烧、精神沉郁和厌食等症状 ,仅引起轻度的鼻炎、中度的咽炎 ,剖检肺表现正常 ,仅见支气管淋巴结肿大 ,45代时无临床症状 ,剖检除扁桃体肿大外没有见到其它症状 ,6 0代毒已完全失去致病力 ,未见任何临床症状及病理变化。
