Tubulointerstitial nephritis (TIN) was established in Wistar rats by direct intrarenal inoculation of TH protein. Expression of T lymphocyte phenotype from the peripheral blood found there were no significant difference in the percentage of CD + 4,CD + 8,CD + 8-CD + 25 ,CD + 8-Ia the ratio of CD 4/CD 8 between the TIN group and the control group at the 7th day after intrarenal inoculation (P<0.05). But the expression of CD 4.CD 8/CD 25 ,CD 8-Ia and CD 4/CD 8 of the TIN group were higher than that of the control group at the 14th day,which indicate that the Tlymphocyte immune response has been happened. These results showed that the system of cellular immunity has been indeed activated in the pathogenetic mechanism of TIN induced by THP. It resulted in the damage of renal tissue. 采用Tamm-Horsfal蛋白(THP)直接肾内注射建立大鼠的小管间质性肾炎(TIN)模型,TIN组9只动物均发生小管间质性组织病理损伤,局灶性单核、淋巴细胞炎症细胞浸润;对照组仅1只轻度炎症改变。外周血T淋巴细胞免疫表型分析,术后7dTIN组与对照组间CD+4、CD+8、CD+8-CD+25、CD+8-Ia+抗原的表达及CD4/CD8的比值均无显著性差异(P>0.05),而术后14d、TIN组均见升高,与对照组比差异有显著性(P<0.05),表明TIN组出现T淋巴细胞的免疫应答反应和活化性细胞毒T细胞明显增加,从而证实THP在致TIN免疫发病机制中,确实存在免疫系统激活,导致肾间质炎症损伤 Objective:The curative effect of IL 1ra on lupus nephritis in NZB/WF 1 mice was studied.Methods:Twelve female NZB/WF 1 mice of 13 weeks (wight 30 g~35 g) were divided randomly into 2 groups.Each mouse in the treated group was intraperitoneally injected with IL 1ra once per 2 weeks for 3 times by dosage of 100 μg (0 1 ml) per time.While in the control group injected with 0 1 ml normal saline.All the mice were killed at the age of 9 months.The kidneys were dissected and fixed with 10% neutral formalin solution... Objective:The curative effect of IL 1ra on lupus nephritis in NZB/WF 1 mice was studied.Methods:Twelve female NZB/WF 1 mice of 13 weeks (wight 30 g~35 g) were divided randomly into 2 groups.Each mouse in the treated group was intraperitoneally injected with IL 1ra once per 2 weeks for 3 times by dosage of 100 μg (0 1 ml) per time.While in the control group injected with 0 1 ml normal saline.All the mice were killed at the age of 9 months.The kidneys were dissected and fixed with 10% neutral formalin solution and embedded in paraffin.The sections were stained with HE, PAS,PASM and Masson,and then observed under light microscope respectively.Results:The control group showed moderate and severe diffuse mesangial proliferative GN with multiple characterisitcs of pathological changes.The treated group showed mild mesangioproliferative GN with simple characterisitc of pathological change in glomeruli,and the degeneration in tubules of kidneys and the inflammation in matrix of kidneys were present permanently.Conclusion:IL 1ra can obviously relieve and improve the renal damage of lupus nephritis in NZB/WF 1 mice. 目的:探讨白细胞介素1(IL1)受体拮抗剂IL1ra对NZB/WF1小鼠狼疮肾炎的治疗作用。方法:同批13周龄,体重30g~35gNZB/WF1雌鼠12只,随机分为二组,治疗组每只小鼠每次腹腔内注射IL1ra100μg(01ml),每2周1次,共3次,对照组每只每次腹腔内注射生理盐水01ml,9月龄时活杀,取肾脏、石蜡包埋制片,作HE、PAS、PASM及Mason三色染色,光镜观察其病理形态学变化。结果:对照组,主要为中至重度弥漫性系膜增生性肾小球肾炎并伴有病变多样性特征,IL1ra治疗组,表现为轻度系膜增生肾小球肾炎,肾小球病变较单一,但肾小管变性及肾间质炎症仍持续存在。结论:IL1ra对狼疮肾炎有明显减轻和改善肾小球损害作用 Objective To investigate the characteristics of the infiltration of the inflammatory cells in the development of chronic interstitial nephritis.Methods PAN was injected subcutaneously to famale Wister rats to induce animal models of chronic intersitial nephritis.Proteinuria per 24 hours respectively in 0,3,5,8,and 12 weeks after the injection was measured.Kidneys in divided groups were taken out respectively,stained with HE and labeled the monoclonal antibodies such as CD4,CD8,MΦ,ICAM 1,CD11a,CD11b,CD18,HA,TCR(α,β),MHCⅡ... Objective To investigate the characteristics of the infiltration of the inflammatory cells in the development of chronic interstitial nephritis.Methods PAN was injected subcutaneously to famale Wister rats to induce animal models of chronic intersitial nephritis.Proteinuria per 24 hours respectively in 0,3,5,8,and 12 weeks after the injection was measured.Kidneys in divided groups were taken out respectively,stained with HE and labeled the monoclonal antibodies such as CD4,CD8,MΦ,ICAM 1,CD11a,CD11b,CD18,HA,TCR(α,β),MHCⅡ with the immunohistochemical ABC technique.Resuls From the 3rd week after injection,the proteinuria per 24 hours increased gradually in experimental group.There was a significant difference compared with the control group.The infiltration of the inflammatory cells was found in renal interstitium by HE staining.The CD4,TCR and ICAM 1 positive cells incrcased at the same time.In the 8th week,the proteinuria per 24 hours reached the peak level.The interstitial fibrosis and the tubular atrophy appeared.ICAM 1,TCR,CD44,HA,CD11a,CD11b,CD18 and MHC Ⅱ positive cells reached the peak level.In the 12th week,the expression of varieties of cytokines and the quantity of the protein tended to decrease.Conclusion The infiltration and proliferation of CD4 positive cells may initiate the expression of varieties of cytokines which plays a key role in the development of chronic interstitial nephritis. 目的 探讨慢性间质性肾炎发生发展中炎症细胞浸润与粘附因子表达的特点。方法 用雌性Wister 大鼠,皮下注射嘌呤霉素氨基核苷(PAN) 建立慢性间质性肾炎动物模型。于注射后0 、3 、5 、8、12 周分别检测24 小时尿蛋白定量,分批取出肾脏作HE染色,免疫组织化学ABC法标记单克隆抗体CD4、CD8、MΦ、粘附分子1(ICAM1) 、CD11a、CD11b、CD18、CD44 、HA、TCR(α,β)、MHCⅡ。结果 实验组第3 周始24 小时尿蛋白量逐渐增加,与对照组比较有显著性差异,HE染色可见肾间质炎症细胞浸润,CD4 阳性、TCR阳性、ICAM1 阳性细胞增多,第8 周24 小时尿蛋白量达高峰,出现肾间质纤维化,肾小管萎缩、ICAM1、TCR、CD44、HA、CD11a、CD11b、CD18、MHC Ⅱ阳性细胞数达高峰;第12 周,多种细胞因子表达及24 小时尿蛋白定量呈下降趋势。结论 CD4 阳性细胞的浸润增殖,启动了多种细胞因子表达,在慢性间质肾炎发生发展中起重要作用。
