Different doses of testosterone propionate(TP) were instituted to treat the transplantable tumors of androgen-independent prostate cancer cell line (PC3m)in nude mice so as to observe the growth of transplantable tumors and changes of adenylate cyclase (AC)and guanylyl cyclase (GC ) activities in tumors.The growth of the transplantable tumors was found to present the“biphasic effects" with concurrent changes in AC activity. The above result might provide some information for the use of high dose TP in the treatment of relapse prostate cancer. The correlation between the suppressive effect of high dose androgen and the AC-GC activities were discussed.

为研究VP-16对雄激素非依赖性前列腺癌细胞PC-3凋亡的诱导作用,应用HE染色观察细胞的形态学改变,以DNA凝胶电泳和流式细胞术(FCM)分析细胞凋亡和细胞周期分布,同时应用3H-TdR掺入法研究VP-16对PC-3细胞DNA合成的影响。结果表明,(1)凋亡的PC-3细胞呈明显的形态学改变;(2)DNA断裂呈梯形变化;(3)PC-3细胞凋亡率随VP-16作用浓度增加和时间延长逐渐增高;(4)VP-16能够明显抑制PC-3细胞DNA的合成。提示VP-16可诱导前列腺肿瘤细胞凋亡。

Purpose:To study the chemosensitivity in human androgen independent prostate cancer cell. Methods: Using the MTT method to study the chemosensitivities to six chemical drugs in human androgen independent prostate cancer cell (PC 3M).Results: The chemosensitivities to six chemical drugs in PC 3M are camptothecin>etoposidum>carboplatin,doxorubincin,nitrogen mastrad>5 fluorouracil (P< 0.05 ).Conclusions: PC 3M is more sensitive to camptothecin,etoposidum, this may give reference to clinical treatment.

目的 :研究人雄激素非依赖性前列腺癌细胞对化疗药物的敏感性。方法 :采用噻唑蓝 (MTT)法测定人雄激素非依赖性高转移前列腺癌细胞 (PC- 3M)对 6种化疗药物的敏感性。结果 :PC- 3M对 6种化疗药物的敏感性依次为喜树碱 >依托泊甙 >卡铂、阿霉素、氮芥 >5 -氟尿嘧啶 (P均 <0 .0 5 )。结论 :PC- 3M对喜树碱、依托泊甙较敏感。可作为临床用药参考。
