本文用~3H-TdR 及放射自显影法测定小鼠可移植性前胃鳞状细胞癌(GS-742)的细胞周期和有关参数。实验结果:GS-742细胞增殖周期:Tc 为20小时,TG_2为2.5小时,TS 为13小时,TG_1为3.5小时,TM 约为1小时。GS-742细胞动力周期各参数分别为:细胞标记率(LI)为41.50±1.71,有丝分裂率(MI)为3.23%±0.23,标记有丝分裂率(LMI)为8.00%±1.08。生长指数为63.85%。这对于采用本模型进行实验研究时合理使用时相特异抗癌药物有一定参考意义。

It was Jound that the proliferative cycle of the young cells in the intestinal crypts which were severely injuried by irradiation was significantly retarded, and their G1-phase to G2-phase were prolonged. These cells gradually changed into misshapen cells. Some of the misshapen cells could not divide and finally they wer.e desquamated; abortive proliferation was found in some of them; some others showed pathological karyokinesis. Thus polymorphic cells were formed. It was difficult to show that the misshapen cells could take part in the repair of the intestinal epithelium. It is speculated that the misshapen cells might to some extent have the function of absorption, secretion and barrier.

140只小鼠,分为单放(~(60)Co 12Gy)、放烧(12Gy+15%体表皮肤Ⅲ度烧伤)和正常对照组。观察小肠上皮畸形细胞的形成及转归。认为小肠隐窝受射线损伤较重的幼稚细胞增殖周期受到严重阻滞,G_1期到G_2期均延长逐渐形成畸形细胞。有的畸形细胞不分裂直到脱落;有的发生流产性增殖;有的则发生病理性核分裂,从而形成形态多样化细胞。难以证实畸形细胞能起到修复肠上皮的作用,但它可能具有一定的吸收、分泌功能,并起到一定的屏障作用。

The authors have determined the effects of 261-B_2-3 and C_(12)b_3-2, extracts two strains of Alternaria alternata, on the nucleolus organizer regions (NORs) activity and the progression of cell proliferating cycle of human peripheral lymphocytes. The methods used in the experiment were selective silver staining of human NORs and cytophotometry. The result showed that the number of chromosomes bearing Ag-NORs was meaningly decreased when the cells were treated with different concentrations of 261-B_2-3 and C_(12)-b_3-2...

The methods used in the experiment were selective silver staining of human NORs and cytophotometry. The result showed that the number of chromosomes bearing Ag-NORs was meaningly decreased when the cells were treated with different concentrations of 261-B_2-3 and C_(12)-b_3-2 extracts. The difference was quite significant when the data were compared with those of the control (P<0.01). It indicated that extracts of both stracts were able to suppress NORs activity of lymphocytes obviously. After the cells had been exposed to C_(12)b_3-2 extracts of different concentration, G_1% increased distinctly and S%, G_2+M% reduced significantly. The difference of data between the control and the treated group was very significant (P<0.01). It suggests therefore that C_(12)b_3-2 possibly acts at late-G_1 phase or carly-S phase and results in delay and block of the progression of cell cycle.

本实验应用人类核仁形成区(NORs)选择性银染方法和细胞光度术,观察了互隔交链孢霉两菌株提取物261-B_2-3和C_(12)b_3-2对人淋巴细胞NORs活性的影响以及后者对细胞增殖周期移行的影响。结果显示,应用不同浓度的261-B_2-3和C_(12)b_3-2提取物处理细胞后,细胞携带Ag-NORs的染色体数明显减少,与对照组比较差异极显著(P<0.01)。表明两株提取物能明显抑制淋巴细胞NORs的活性。细胞暴露于不同浓度的C_(12)b_3-2提取物后,G_1%明显增多,S%和G_2+M%显著减少,与对照相比差异极显著(P<0.01)。说明C_(12)b_3-2提取物可能作用于G_1后期或S早期,延缓和阻滞了细胞周期的移行。
