The experimental conditions were optimized including the method of RNA extraction,primer concentrations in cDNA synthesis and first-stage PCR,reaction volume of PCR and temperature cycling protocols.The“nested”PCR detected HCV RNA in 52(78%)of 67 sera from patients with non-A,non-B hepatitis.It was also found that HCV RNA was detected in 16(59%)of 27 patients with non-A,non-B hepatitis without detectable antibodies to HCV by commercial assay kit.These findings confirm that HCV is an important cause of non-A, non-B hepatitis and suggest that the prevalence of HCV infection may be underestimated by detecting antibodies to HCV only.

分析126例丙型肝炎病者的临床资料,丙型肝炎病毒血清抗体(抗-HCV)滴度及丙型肝炎核酸(HCV-RNA)。发现有明确输血(浆)史者103例,该病史中发现输血后2个月内有肝损者87例占84.4%。抗-HCV检出率为91.3%,其中急性病者检出率82.6%,慢性者95.8%。抗-HCV强阳性者(OD>1.0),HCV-RNA的检出率48.1%,明显高过OD<1.0的低阳性者。抗-HCV阴性者中有18·18%(2/11)检出HCV-RNA。另外,HCV-RNA的检出与谷-丙转氨酶(ALT)水平有一定关系,有肝损者比ALT正常者HCV-RNA检出率明显高。提示HCV-RNA的检出与活动性肝病有关系。
