Enantioselective extraction of terbutaline enantiomers by lipophilic tartaric acid
      
Distribution behavior of terbutaline enantiomers was examined in the aqueous and organic solvent of a two-phase system containing L-dibenzoyltartaric acid and lipophilic phase transfer reagent of Na-tetraphenylborate.
      
The influences of pH, organic solvents, concentrations of Na-tetraphenylborate and L-dibenzoyltartaric acid on the partition coefficients and enantioselectivity of terbutaline enantiomers, were investigated.
      
The results show that tetraphenylborate lipophilic anion and terbutaline enantiomers form two lipophilic salt complexes, which facilitates the solubility of the enantiomers in the organic phase.

Objective: To investigate the factors infleuncing the separation of terbutaline enantiomers by capillary zone electrophoresis(CZE) using β cyclodextrin derivatives as chiral additives.Methods: Analyses were carried out on an Agilent 3D CE capillary electrophoresis system equipped with a diode array detector.An untreated fused silica capillary (58.5 cm×50 μm,50 cm effective length) was used.The voltage was 20 kV and the temperature was 20℃.The detective wavelength was set at 204 nm.Samples were injected into the capillary by 5 kPa pressure for 3 s.Results: Baseline separation of terbutaline was achieved when using 2,3,6 trimethyl β cyclodextrin(TM β CD) as chiral additive but not when using carbomethyl β cyclodextrin(CM β CD).When using TM β CD,the separation rate increased with the decrease of pH with obvious changes within 2.5 4.0. There was no obvious changes of separation rate when using CM β CD,and when pH>5.5 the separation was not achieved.The compound of cyclodextrin and terbutaline increased with the increase of cyclodextrin when the volume rate of cyclodextrin was 0.4% 1.6%,making terbutaline easier for separation.Conclusion:The types of β CD,the concentration and pH of buffer are the major factors influencing the separation of terbutaline and it can be completely separated.

目的 :考察毛细管区带电泳法 (CZE)中各种因素对β-环糊精衍生物分离肾上腺素类药物特布他林的影响。方法 :采用 CZE对特布他林的手性分离进行研究。未涂渍熔融石英毛细管 5 0μm× 5 8.5 cm (有效长度 5 0 cm ) ;温度 2 0℃ ,操作电压 2 0k V,正极进样 5 k Pa× 3s,检测波长 2 0 4 nm。β-环糊精的两种衍生物作为手性添加剂。结果 :2 ,3,6 -三甲基 -β-环糊精为手性选择剂时使特布他林得到了基线分离 ,羧甲基 -β-环糊精为手性选择剂时却不能得到基线分离。特布他林在三甲基 -β-环糊精环境下 ,分离度随 p H变小逐渐增大 ,p H在 2 .5~ 4 .0范围内变化明显 ;而在羧甲基 -β-环糊精环境下分离度随 p H变化不明显 ,且当 p H>5 .5时特布他林不能分离。环糊精体积分数从 0 .4 %~ 1.6 %时 ,环糊精与特布他林形成的复合体随环糊精的增加而增加 ,使该手性物质更容易拆分。结论 :β-环糊精的类型、浓度以及缓冲液的 p H是影响特布他林手性拆分的主要因素 ;肾上腺素类药物特布他林能得到完全分离

Chiral separation of terbutaline by RP-HPLC with a new chiral column Shiseido CD-Ph has been studied. The effects of the content of acetonitrile, pH, column temperature and buffer concentration on the chiral separation of terbutaline were discussed. At the optimal conditions, baseline separation of terbutaline was achieved. The results achieved indicated that the method was reliable, simple, rapid and sensitive and could be applied to routine analysis of enantiomers of terbutaline.

采用新型商品手性柱ShiseidoCD Ph,在反相条件下,对β受体阻滞剂类药物特布他林进行了手性拆分研究。考察了流动相组成、pH、柱温及缓冲盐浓度对特布它林手性拆分的影响。在优化的实验条件下,该药物获得基线分离。该方法可用于该类药物的常规分析。
