Objective Fluorescent-labeled antisense oligomer that is hybridized to 18-bp sequences next to the start coden of human proliferating cell nucleus antigen (PCNA) gene was used to assess the intracellular distribution and stability of the oligonucleotides in the presence or absence of liposome (lipofectin). Method IUM 18-mer Phosphorothioate and unmodified antisense oligonucleotides conjugated with fluorescent 5' -isothiocyananate (5 '-FITC) were cncapsulated by lipofctin or without, and then added into human bladder cancer cell BIU-87 culture media. The celltular distribution was observed by fluorescence microcopy in fixed cells. Result In the abscnce of lipofectin, the oligonucleotide localized to discrete structures in the cytoplasm of the cells, resulting in a punctate fluorescence pattern . In the presence of lipofectin, cellular fluorescence was markedly increased and the oligonucleotide localized with the nucleus, as well as to discrete stuctures in the cytoplasm. Accumulation of the oligonucleotide in the presence in the presence of lipofectin was time -dependent. Conclusion Cationic lipids can increase the amount of oligonucleotide associated with cells and alter the intracellular distribution of the oligonucleotide .The phosphorotide. The phosphorothioate antisense oligonucleotide remained in the nuclei as well as cytoplasm more stable than unmodified .

目的观察硫代修饰型及天然型PCNA反义寡核苷酸在脂质体介导下或直接转染人膀胱癌细胞BIU-87后在细胞内的分布及其稳定性。方法将异疏氰酸荧光素(5'-FITC)标记的18mer硫代磷酸化修饰型及未修饰型PCNA反义寡核苷酸在脂质体介导下或直接转染人膀胱癌细胞BIU-87,应用荧光显微镜观察转染细胞从细胞内的时相分布。结果修饰型反义核酸直接转染细胞后30分钟,少数细胞胞浆荧光呈离散型、点状分布,4小时后有少数细胞胞核有强荧光聚积。在脂质体介导下,荧光细胞数目明显增加,4小时后绝大多数细胞迅速积聚于细胞核,4~8小时后,细胞核内荧光强度进一步增强,12小时后细胞荧光减弱甚至消失。而非修饰型反义核酸在直接转染或脂质体介导下均发现荧光在3小时后消失。结论脂质体增加PCNA修饰型反义寡核苷酸与膀胱癌细胞BIU-87结合的数量,同时使其在细胞核内分布聚积明显;PCNA反义寡核苷酸硫代修饰具有更好的稳定性。

Objective To observe the ordered growth behavior of human urinary bladder cancer cell line under culture in vitro . Methods The suspension of human urinary bladder cancer cells was inoculated to the localized areas. After the cells grew alone the wall to form colonies, macroscopic and microscopic observations of the spreading diameter, spreading rate, cell shape, average cell density, average cell size and dehydrogenase activity and its sensitivity to pH were dynamically performed. Results Under culture conditions, obvious laminar structure appeared in the locally growing colonies of human urinary bladder cancer cells and there was difference of the shape, size, density and dehydrogenase activity and its sensitivity to pH among the cells in different zones. Conclusion Space closing and biodissipation lead to the self organization of the human urinary bladder cancer cells. Consequently they show ordered growth behavior. In this paper, a simple and controlled model of wavy growth of human cells is provided.

目的 观察人膀胱癌细胞 (BIU)在体外培养条件下的生长有序行为。方法 将BIU细胞悬液进行局限区域接种 ,待其形成贴壁生长的群落后进行宏观和显微动态观察及参数测量 (扩展直径、扩展速率、细胞形态、细胞平均密度、细胞平均大小、脱氢酶活性、对pH的敏感性等 )。结果 体外细胞培养条件下 ,局限区域生长的BIU细胞群落能出现明显的层次特性 ,不同区域细胞的形态、大小、密度、脱氢酶活性以及对pH的敏感性均存在差异。结论 空间封闭和生物耗散导致细胞自组织 ,其结果表现为生长有序行为。本实验为人体细胞的波动生长提供了一个简单可控的初步模型

Hybridization in situ was used to detect proliferating cell nuclear antigen (PCNA) mRNA expression of BIU-87 cells which were treated with antisense oligodeoxynucleotides (ASODN) conjugated with lipofection. The results showed that PCNA mRNA expression of BIU-87 cells was significantly suppressed or reduced in comparison with the controls. Hybridization in situ was used to detect proliferating cell nuclear antigen (PCNA) mRNA expression of BIU-87 cells which were treated with antisense oligodeoxynucleotides (ASODN) conjugated with lipofection. The results showed that PCNA mRNA expression of BIU-87 cells was significantly suppressed or reduced in comparison with the controls. But the SODN showed no such effect in comparison with the controls. It was concluded that ASODN can obviously inhibit the proliferation of bladder cancer cells by inhibiting the expression of PCNA.

运用原位杂交技术检测脂质体介导增殖细胞核抗原 ( PCNA)反义寡脱氧核苷酸抑制人膀胱癌细胞 BIU- 87PCNA m RNA的表达水平。结果表明脂质体介导 PCNA反义寡脱氧核苷酸处理组中 BIU- 87细胞 PCNA m RNA的表达水平明显低于对照组。结果提示脂质体介导 PCNA反义寡脱氧核苷酸可抑制人膀胱癌细胞 BIU- 87体外增殖活性 ,其抑制作用可通过阻断 PCNA蛋白表达来实现。
