The effect of a low midpoint redox potential cytochrome c_3 on PSP of chroma-tophores from Rhodopeudomonas capulata has been investigated.The restoration of cyclic (endogenous) and non-cyclic (with DCPIPH_2 asan electron donor and vitamin k_3 or fumarate as an electron acceptor) activityof PSP of ferredoxin-depleted chromatophores can be promoted by the addition of cytochrome c_3.Under a gas phase of Ar,the optimum concentration of cytochromec_3 for the stimulation of cyclic PSP activity in normal chromatophores is 1.8 μmol.In the presence of PMS (10~(-4)mol/L), the stimulation rises linearly with the increaseof cytochrome c_3 concentration and then becomes constant when certain value hasbeen approached.The cytochrome c_3-involved PSP activity is inhibited by antimycin A (10~(-7)mol/L), but the inhibition disappears when PMS is added simultaneously. Also,Jow concentration of o-phenanthroline (10~(-5)mol/L) has an inhibitive action onthe cytochrome c_3-involved PSP. Adding PMS at the same time can eliminateits inhibitory effect, but not if the concentration of o-phenanthroline is raised to10~(-3) mol/L.Under a gas phase of H_2, the PSP activity is less than the one under Ar anddecreased rapidly by freezing 60% of the activity being lost after 24 hrs at-20℃.The activity can be stimulated by adding c_3, but not ferredoxin. The PSP activityis increased further by joint addition of both.The possible role of cytochrome c_3 in electron transport of hydrogen metabolismof Rhodopseudomonas capsulata has been discussed.

We discovered that the PBC of the IE is related not only to the acid-base property of the carriers, but also to those of the enzymes.If the carrier has opposite electric charge to that of the enzyme, the PBC and the thermal stability of the IE will increase, if the carrier and enzyme have the same kind of electric charge, the PBC of the IE either remaing unchange or decreases. We also discovered that when the porosity of the ionic carriers and the concentration of the buffer were constant, if the enzyme was only coupled to the carriers'outer surface, the IE's optimal pH would not change; and if most of the enzyme was coupled to the carriers' internal surface, the IE's optimal pH would probably change. We noted that after the carboxyl groups and amino groups were introduced to Sephadex, the resistance of the carrier to microbial contamination was largely increased.

Trypsin was coupled to porous triethanolaminepolystyrene (TMPS) resin by CNBr. IR analysis indicated that the reaction mechanism of coupling enzyme to TMPS is similar to that of coupling enzyme to polysaccharide by CNBr. Trypsin was coupled to porous triethanolaminepolystyrene (TMPS) resin by CNBr. IR analysis indicated that the reaction mechanism of coupling enzyme to TMPS is similar to that of coupling enzyme to polysaccharide by CNBr. The immobilized trypsin increased with the increase of the CNBr concentration but its specific activity decreased.The amount of immobilized trypsin was appropriate and the specific activity was retained relatively high at coupling pH of about 10. Since trypsin-catalyzed reaction involved the liberation of hydrogen ions, the hydrogen ion concentration in the microenviroment is higher than in the bulk solution. Consequently, the pH optima of immobilized trypsin shifted to basic region and broadened.Trypsin retained only 48% of its enzyme activity after heating at 60℃ for 90 min. In contrast, under the same condition the immobilized trypsin showed enhanced resistance to heat inactivation, retaining 90% of its original activity. Because of low cost and good mechanical strength, TMPS may become a very useful matrix for covalenting enzymes.
