For removing protein, the lysate was extracted with phenol-chloroform, followed by adding sufficient NaCl (5M NaCl) to bring the solution to 0.6M...

For removing protein, the lysate was extracted with phenol-chloroform, followed by adding sufficient NaCl (5M NaCl) to bring the solution to 0.6M NaCl, then put the lysate on ice for two hours. Most of the proteins and the denatured chromosome DNA were thus removed and the tubid aqueous DNA solution turned to be clear. This technique is also proved to be useful in isolating small plasmid from a variety of other bucteria.

The results show that the content of C_(org) is from 0.5% to 2.89% in fine grained or carbona- ceous sediments, and 0.5% to 1.51% in the diatomites, the chloroform extra- cts are from 835.1ppm to 3901.5ppm; the saturated hydrocarbons fraction of extracts are composed mainly of normal alkanes, most of them are nC_(23)-- nC_(33), dominated by C_(29) or nC_(31) with remarkable odd carbon predominance (OEP≥4). Some terpenoid and steroid hydrocarbons were identified and the content of terpenoids was measured to be much higher than that of ste- roids in the diatomite sediments. Terpenoids identified consist mainly of C_(27)--17β(H) hopane, C_(29)--17β(H), 21α(H) hopane, C_(29)--17β(H), 21β(H)ho- pane, C_(30)--17β(H), 21β(H) hopane, C_(30)--17α(H), 21β(H) hopane, C_(31)-- 17β(H), 21β(H) hopane, C_(27)--hopene and C_(30)--hopene. Another typical com- pound deteted in large amounts is de-A-Lupane of high plant origin. By comparison, The composition and distribution of biomarker compounds in all the diatomite samples studied are quite similar but different from those in other types of sediments from the Tengchong Basin, yunnan Province.
