To establish a protocol for isolating,culturing,proliferating urothelial cells,and to obtain large number of autologous urothelial cells for tissue_engineered substitutes.Urothelial cells were isolated from sample tissues by scraping or tissue explanting and were cultured in Karetinocyte_SFM.Morphology and expansion of the cells were observed.Cell_type specific proteins were analyzed by immunohistochemical methods.The results showed that urothelial cells presented a typical morphological feature of transitional epithelium.Cells could be passaged 10 times without a noticeable decrease in rate of cell proliferation.Immunohistochemistry showed that CKAE1/AE3 was positive and CK34βE12 was negative.Therefore,in this protocol,the isolation of urothelial cells are simple,the cells proliferate rapidly and can be passaged easily.The biological properties of urothelial cells have been kept and no signs of senescence were seen even after the 10th passage.

Methods 101 cases of urinary calculi treated by the frequency-doubled double-Pulse U-100 Nd of YAG laser under ureteroscope and cystoscope were reviewed retrospectively, included 83 cases with ureterolith, 15 with cystolith and 3 with posterior urethral calculus. Results The successful rate of lithotripsy was 91.6%(76/83) in ureterolith, 80%(12/15)...

Objective To evaluate the effect and safety of the frequency-doubled double-pulse U-100 neodymium of YAG (Nd:YAG) laser lithotripsy of calculi in the urinary tract. Methods 101 cases of urinary calculi treated by the frequency-doubled double-Pulse U-100 Nd of YAG laser under ureteroscope and cystoscope were reviewed retrospectively, included 83 cases with ureterolith, 15 with cystolith and 3 with posterior urethral calculus. Results The successful rate of lithotripsy was 91.6%(76/83) in ureterolith, 80%(12/15) in cystolith, and 100%(3/3) in posterior urethral calculus. The mean time of operation was 33 minutes and the laser worked 5.5±2.8 minutes in average. The stonefreeing time was 7~28 days and the average time was 15 days. Conclusions The frequency-doubled double-pulse U-100 Nd of YAG laser can shatter stones quickly and efficiently with no damage to the normal urinary organ.

目的 探讨双频双脉冲U 10 0掺钕钇铝石榴石 (Nd :YAG )激光粉碎泌尿系结石的疗效及安全性。 方法 应用双频双脉冲U 10 0Nd :YAG激光经输尿管镜和尿道膀胱镜治疗泌尿系结石10 1例 ,其中输尿管结石 83例 ,膀胱结石 15例 ,后尿道结石 3例。 结果 输尿管结石碎石成功率为91.6 % (76 /83) ,膀胱结石碎石成功率为 80 .0 % (12 /15 ) ,尿道结石碎石成功率为 10 0 % (3/3)。平均手术时间 33min ,平均激光工作时间 (5 .5± 2 .8)min。输尿管结石碎石后平均 15 (7~ 2 8)d排净结石。 结论 双频双脉冲U 10 0Nd :YAG激光能迅速、高效地粉碎泌尿系结石 ,对正常泌尿系器官无损伤
