Objective: To construct a human gallbladder carcinoma cell line and to investigate its biological behavior. Methods: GBC-SD, a human gallbladder carcinoma cell line was constructed via the tissue culture. The biological behavior such as cell morphology, cytokinetics, CEA and CA-199 secretion and growth pattern in heteroimplantation were studied in vitro and in vivo respectively. Results: The GBC-SD cells were mainly composed of square, polygonal and spindal cells, the doubling time was 21.4h. The cell line could secret CEA and CA-199 continuously. The histological features of implanted tumor in nude mice were similar to those of the original one. Conclusions: Being a new constructed gallbladder carcinoma cell line, the GBC-SD cell line could be served as an experimental model for further study.

Objective To study the mechanisms of multidrug resistance (MDR) in human gallbladder carcinoma cell line and establish the MDR cell model of human gallbladder carcinoma.Methods The amplification of MDR genes (MDR1,MRP and LRP) in human gallbladder carcinoma cell line BGC-SD was detected by using reverse transcript polymerase chain reaction (RT-PCR).The immunohistochemistry was performed to detect the expression levels of P-gp,MRP and LRP.The flow cytometry (FCM) was carried out to determine the expression levels of P-gp,GST-π and TOPOII.The IC50 of 8 anticancer drugs to GBC-SD cells was determined by MTT method.Results The amplification of MDR genes (MDR1,MRP,LRP) and the expression of P-gp,MRP and LRP were positive in GBC-SD cells.There was no significant difference in the expression levels of GST-π and TOPOII between the GBC-SD cells and control group.GBC-SD cells were less sensitive to 8 anticancer drugs.Conclusion With the amplification of the genes MDR1,MRP and LRP,GBC-SD is an intrinsic MDR cell line.It can be used as an optimal MDR cell model for further study of multidrug resistance of gallbladder carcinoma.

目的 探讨人胆囊癌细胞系GBC SD耐药性产生的机制 ,建立先天性耐药的人胆囊癌细胞模型。方法 用逆转录 聚合酶链反应 (RT PCR )检测GBC SD细胞多药耐药相关蛋白(MDR1)、多药耐药相关蛋白 (MRP)和肺耐药蛋白 (LRP)基因扩增 ;免疫细胞化学染色检测该细胞系P 糖蛋白 (P gp)、MRP和LRP蛋白表达 ;流式细胞术测定P gp、谷胱甘肽 S 转移酶 (GST π)和DNA拓扑异构酶Ⅱ (TOPOⅡ )表达水平 ;用四甲基偶氮唑蓝 (MTT)比色法检测该细胞对 8种抗癌药物的敏感性。结果 人胆囊癌GBC SD细胞中MDR1、MRP和LRP基因呈阳性扩增 ,P gp、MRP、LRP蛋白表达阳性 ;GST π和TOPOⅡ的表达水平与对照组细胞差异无显著性。四唑盐比色结果显示 ,GBC SD细胞对 8种抗肿瘤药物均有程度不同的耐受性 ,耐药倍数从 5 .3倍到 42倍不等。结论 人胆囊癌细胞系GBC SD是一株先天性耐药的恶性肿瘤细胞系 ,其耐药性的产生与MDR1、MRP、LRP基因扩增有关。

Objective:To establish a novel subcutaneous tumor model in nude mice by using hu-man gallbladder carcinoma cell line GBC-SD and observe its biological characteristics.Methods:GBC-SD cell line was cultured and the cultured cells were harvested to make cell suspension.Each of30mice was injected0.2ml GBC-SD cell suspension(2×10 6 cells)subcutaneously into femoral outside.Then tumor forma-tion rate,tumor appearing time,growing characteristics and its effects on general condition of the nude mice were observed,and anatomical and pathological examination was performed as well.Results:The overall tumor formation rate was100%,and the average tumor appearing time was1week after tumor cell suspension injection.Cachexia occurred in about the8th week,and the average life-span of the mice was about 20weeks.Con clusion:The human gallbladder carcinoma subcutaneous tumor model was suc-cessfully established and can now be used in the researches concerning gallbladder carcinoma treatment.

目的:建立人胆囊癌裸鼠皮下瘤模型,并对其生物学特性进行初步观察。方法:培养人胆囊癌细胞系GBC-SD细胞,收集细胞并制备细胞悬液。每只裸鼠于股外侧皮下注射GBC-SD细胞悬液0.2ml(约2×106个细胞),共30只。观察GBC-SD细胞系成瘤率、成瘤时间、肿瘤生长情况及对裸鼠机体状况的影响。解剖观察瘤体情况并行病理切片观察。结果:裸鼠成瘤率为100%,GBC-SD细胞系对高周龄裸鼠亦有致瘤性,成瘤时间为1周左右。裸鼠于注射瘤细胞后约8周出现恶液质,晚期恶液质明显并衰竭死亡。荷瘤生存期平均为20周。结论:GBC-SD人胆囊癌裸鼠皮下瘤模型成瘤率高,GBC-SD细胞系对高周龄裸鼠亦有致瘤性。肿瘤生长速度稳定,易于观察,可作为胆囊癌治疗等研究的良好平台。
