In the present work, we studied immunosuppressive factors produced by several histologically distinct tumor cell lines. They were referred to as tumor-derived immunosuppressive factos (TDSP). The TDSP inhibited mouse spleen cell response to concanavalin A and alloantigens. They also inhibited interleu-kin 2 (IL-2) production, IL-2 responsiveness and IL-2 receptor expression. To study the correlation between the Jaffe grading system of giant cell tumors of bone and tumor cells proliferation.Methods Twenty seven cases of giant cell tumor of bone were evaluated for their cell kinetics by using proliferating cell nuclear antigen(PCNA)immunoreactivity.Results The multinucleated giant cells nuclei were unlabeled by PCNA and the major proliferating cells in the tumors were identified as mononuclear stromal cells. No significant correlation existed between the PCNA labeling index and histopathological grading of the giant cell tumors.Conclusion The Jaffe grading system does not correspond with the cell proliferation potential in these tumors as assessed by PCNA immunohistochemistry.The multinucleated giant cells do not form a proliferating population and may be not tumor cells.
