The results showed that there was a tendency of equilibrium in the production and elimination of free radical in group I , thus assuring the structural and functional integrity of the cell : as compared with group I , the activity of SOD in group Ⅱ was higher , the number of mitochondria and rough endoplasmic reticulum increased , the glycogen was distributed lo- cally : In the exhausted group , the quantity of MDA in liver cells remarkably increased : the activity of SOD de- creased , the mitochondria in the liver cell reduced , the endoplasmic reticulum broke down and the cells became blank , the structure of cell membrane was damaged . a number of cytoplasm entered the sinus . It was considered that the increased free radical could cause the damage of cell membrane after strenuous exercise , and the dam- age of the structure and function of the mitochondria in liver cell provided the foundation of the producing LPOthat intervened the damage process of the liver tissue , which cause the leak of Yenzyme from the cell . The lipid peroxidation is the principal cause of the increased activity of enzyme in serum . It is important that remarkably increased SOD can prevent the mitochondria and cell from being damaged by free radical , assure the mitochon- dria maintaining a normal breathing and energy supply , and preserve the physiological function of the organ .

本文研究不同时间游泳训练对大鼠肝组织过氧化脂质(LPO)、超氧化物歧化酶(SOD)活性的影响,同时观察肝组织超微结构的变化。结果显示:实验组Ⅰ自由基产生与清除基本达到动态平衡,保证了细胞结构和功能的完整;实验组ⅡSOD活性与实验组Ⅰ相比显著升高,肝细胞内线粒体多,粗面内质网丰富,糖元多呈区域分布,膜结构完整等超微结构的适应性改变,促进了肝组织的物质和能量代谢。力竭组大鼠肝细胞中MDA含量显著增加,SOD活性有所下降,肝细胞内线粒体减少,内质网断裂,细胞空化,膜结构破损,大量内容物进入血窦。认为运动后自由基增多是导致细胞膜结构损伤的因素之一,肝线粒体结构和功能受损,进一步提供产生LPO的基础,UO明显介入肝组织损伤的过程,从而引起细胞内酶外逸,所以脂质过氧化也是引起血清酶活性增高的主要原因。SOD显著增加对防止线粒体及细胞免受自由基损伤,保证线粒体进行正常呼吸供能,进一步维持整个器官的生理功能具有重要意义。

Pulsed electric fields(PEFs) with fixed frequency, width and gradually increased peak value of voltage was applied to 30 healthy rabbit liver tissues.The specific aims were to explore the feasibility of establishing a model of in vivo PEFs distribution in healthy rabbit liver tissues and to provide important references for clinical electrochemotherapy and for electrotransfer.Repeated experiment and self-comparison statistics design were implemented.The rabbit underwent the experiment under intravenous anesthesia and their liver tissues, after exposure to PEFs, were sent for HE staining.Necrotic borderline was visible 3 days after PEFs application,the necrotic shape of concentric circle was evident around the electrodes under optical microscope at lower voltage,as voltage increasing,two necroses in the shape of concentric circle gradually enlarged; nuclei with chromatin condensation,fragmentation and lysis alterations were seen in the middle region between the needles; concentric circles changed into ellipse fusiform and finally overlaped each other forming irregular necrosis contours.Cell cavitation and tissues ischemia were also observed within electric field. The shape of tissue necrosis from the experiment was noted to correlate with theoretic simulation of electric field distribution.Therefore, rabbit liver tissues can be a good carrier for in vivo modeling of electric field distribution when the lethal effects of PEFs in tissues are investigated. PEFs also show safety for the surrounding normal tissue while causing damage or injury to the target area therapeutically.

通过观察频率、脉宽固定,电压峰值递增的脉冲电场对30只新西兰大白兔正常肝脏的杀伤效应,探讨在兔正常肝组织中建立脉冲电场分布模型的可行性,为临床电化学或电转基因治疗提供重要参考。采用重复实验及自身对照,脉冲处理3d后取材HE染色,光镜下见低脉冲剂量时两电极周围呈同心圆形坏死、界限清晰;剂量增大则同心圆形坏死区亦增大,且渐变成椭圆梭形,两电极中点处肝细胞出现核固缩、碎裂、溶解样改变,最后两椭圆梭形坏死相互重叠融合成不规则坏死,处理区尚见肝细胞的空化效应及肝组织缺血现象,实验所见与理论模拟的电场分布能较好吻合。因此,兔肝组织可作为脉冲电场生物组织杀伤效应研究中电场建模的有用载体,电脉冲在有效杀伤靶区组织的同时对周边正常组织是安全的。

目的探讨姜黄素对SW480(人结肠癌细胞)细胞株诱导分化机制。方法应用MTT比色法和流式细胞仪测定细胞周期的变化。并通过电子显微镜观察诱导分化后的亚细胞结构。结果姜黄素对SW480细胞增殖有明显的抑制作用。且呈时间、剂量依赖性,流式细胞仪分析G1期细胞堆积、S期细胞减少、G2/M期细胞增多、亚细胞结构、细胞空化、核仁不规则、染色质趋边凝集。结论姜黄素能够抑制SW480细胞增殖,阻止G1期细胞向S期转化的进程,促进SW480细胞凋亡。
