The activation of random donor platelet concentrates and platelets prepared from random donor apheresis collections (plateletpheresis) during storage were studied. Percentage of CD62p staining and mean channel fluorescence(MCF) of CD41 of two kinds of platelets during storage on day 0, day 1, day 3 and day 5 were determined by flow cytometry. The results showed that percentages of CD62p staining and MCF of CD41 in plateletpheresis were (18 91±6 25)%, (19 48±8 27)%, (22 82±6 06)%, (56 71±11 79)% and (8 09±2 38)%, (8 13±2 45)%, (8 44±2 51)%, (19 87±6 13)%, while the results of platelet concentrates were (30 65±12 33)%, (31 46±11 86)%, (32 51±13 05)%, (63 55±13 27)% and (10 33±4 37)%, (11 09±6 61)%, (13 46±9 69)%, (24 41±10 15)%, respectively. The platelet count and pH value were also determined. The platelet number, pH value, percentage of CD62p staining and MCF of CD41 had no significant difference within 3 days of platelet storage. The platelet number and pH value decreased significantly(P<0 001), while percentages of CD62p staining and MCF of CD41 increased significantly(P<0 001) on day 5 of storage. It is concluded that the quality of plateletpheresis is better than platelet concentrate.

The target endpoints were set at 3.0×10 11 /Lfor single APs(n=22) and 6.0×10 11 /Lfor double APs (n=31). The changes in donor′s platelet counts were determined at pre- and post- donation and one month later. Results The platelet yields for the single and double APs were (2.86±2.39)×10 11 /Land (5.75±4.46)×10 11 /Lrespectively. The changes of platelet counts on donors in pre- and post- donation were significant between ( 211.64 ±21.27)×10 9/Land(135.73±20.94)×10 9/L(P<0.05)forthesingleAPs,between(270.26±37.25)×10 9/L and(141.39±39.28)×10 9/L(P<0.05) for the double(APs),respectively. The platelet counts of donors for the single and double APs in post-donation rates of platelet decreased more than that in pre-donation by 35.0%±12.6% and 47.9%±15.9%, respectively. After donation of one month, the platelet counts of donors for the single and double APs elevated to (215.86±24.56)×10 9/L,(260.32±38.71)×10 9/L respectively, but in double APs the platelet counts were lower in post-donation than that in pre-donation significantly (P<0.05). Conclusion The baseline platelet values of donors for single APs in pre-donation do not influence the quality of platelets and have no adverse effects on the donors′ health. It had been recommend that baseline platelet counts of donors for double APs in pre-donation should be >250×10 9/L, which will reduce the rates of the post-donation values of platelet counts to <100×10 9/L, and increase the recovery rate to the pre-donation values of platelet counts after one month.
