本文采用高选择性固定液涂渍的玻璃毛细管色谱柱分离DNT,TNT异构体。为了寻找选择性高的气相色谱固定液,首先用填充柱涂以典型的固定液,测定DNT,TNT相邻两峰之间的相对保留值(α),从而找到好的固定液。然后把此固定液涂在玻璃毛细管柱上,进行研究。试验结果说明:OV-225毛细管色谱柱适于分离DNT的6个异构体,OV-25毛细管色谱柱适于分离TNT的6个异构体。两者均可达到基线分离而时间又不长。

In this paper, TNT and its five metabolites were separated under the system of 5μm reversed-phase C18 ion-pair column and methanol-water by HPLC. Two diffierent ways were used for hydrolization of metabolites in urine. Hydrolization condition of 4-HA in urine is that urine was acidified with 6N hydrochloric acid(0.1ml/ml urine)and heated at 40℃ for 10 min. Hydrolization condition of other metabolites is that urine was acidified with concentrated hydrochloric acid(0.1 ml/ml urine)and heated at 100℃ for 60 min....

Benzene was selected as the extracted solvent, the ratio of two phase was 1 : 1, pH=7,extracted by shaking 5 min. 本文用高效液相色谱法在5μm反相C~(13)离子对柱、甲醇/水系统下,分离了TNT及五种代谢物。确定了尿样的水解采用两种方式:在40℃下加6N HCl(0.1ml/ml尿)水解10分钟,中和后提取4-HA;在100℃下加浓HCl(0.1ml/ml尿)水解60分钟,中和后提取其它TNT代谢物。提取剂用苯,提取比为1:1,溶液pH=7,振荡时间5分钟。在上述水解条件下可使各代谢物水解量达到最大。
