OBJECTIVE: To summarize and conclude the achievements of collagen applied in the links of bone tissue engineering in recent years, analyze the relationship between structure and function of collagen molecules, expound the great advantages and disadvantages of collagen as a natural biomaterial, and further investigate new approaches to the application of collagen in bone tissue engineering. DATA SOURCES: A computer-based search for the articles related to collagen between January 1995 and December 2004 was performed...

OBJECTIVE: To summarize and conclude the achievements of collagen applied in the links of bone tissue engineering in recent years, analyze the relationship between structure and function of collagen molecules, expound the great advantages and disadvantages of collagen as a natural biomaterial, and further investigate new approaches to the application of collagen in bone tissue engineering. DATA SOURCES: A computer-based search for the articles related to collagen between January 1995 and December 2004 was performed in Medline Database, Chinese Journal of Science and Technology Database(VIP Information Web), China National Knowledge Infrastructure(CNKI) with the key words "tissue engineering, type Ⅰ collagen", and the language was limited to Chinese. STUDY SELECTION: The articles were checked up primarily according to the inclusion criteria: ①studies on bone tissue engineering, ②randomized or blind methods were not excluded, and the exclusion criteria: repetitive studies and reviews. DATA EXTRACTION: Totally 43 related articles to bone tissue engineering were collected, excluding the articles with similar contents, and including the authorized journals published in the last 5 years firstly.Finally 25 articles accordant with the inclusion criteria were analyzed. DATA SYNTHESIS:After the selected articles were analyzed and arranged, the relevant contents to the application of collagen in bone tissue engineering were classified and summarized based on the 3 essential factors for tissue engineering: scaffold, microcarrier and inoculation medium. Besides, type Ⅰ collagen was also an important carrier of cell signals, and had similar function to biological factor. The mechanism of type Ⅰ collagen was concluded although its types were not classified. CONCLUSION: Collagen and its products are widely used in tissue engineering attributing to their good biocompatibility, suitable degradable ability and mild antigenicity, but they are also of intrinsic deficiency. To overcome the lack of natural collagen and produce more new products accordant with the needs of tissue engineering, it is needful to modify and compound collagens.

目的:归纳、总结近年来胶原在骨组织工程各环节中应用所取得的成果,分析胶原分子结构与功能的关系,阐述胶原作为一种天然的生物材料所具备的巨大优势和存在的不足,并进一步探讨胶原在骨组织工程中应用的新途径。资料来源:应用计算机检索Medline数据库、中文科技期刊数据库(维普资讯网)、CNKI数字图书馆1995-01/2004-12相关胶原的文献。检索词“组织工程,I型胶原”,限定文章语言种类为中文。资料选择:对资料进行初审,纳入标准:①与骨组织工程相关文献。②不排除是否为随机、盲法等论证推荐的文章。排除标准:重复研究、综述文献。资料提炼:共收集到43篇与骨组织工程相关的文章。排除其中研究内容相似的文章,以近5年内发表在较权威杂志者优先。对符合标准的25篇文献进行分析。资料综合:通过对入选的文献进行分析、整理,将胶原在骨组织工程中的应用按组织工程三要素相对应的内容进行分类和综合:支架、微载体和接种媒介。另外,I型胶原是一种极其重要的细胞信息载体,具有类似生物因子的作用。虽未对其进行归类,但对其作用机制进行了总结。结论:胶原及其产品以其优良的生物相容性、适宜的可降解性、弱的抗原性在骨组织工程中得到广泛应用,...
