900bp DNA fragment of hepatitis B virus suriaceantigen

为了进一步研究乙型肝炎病毒(HBV)的s基因变异在免疫逃逸中的作用,我们通过酶切消化质粒载体pEcob6(含双拷贝HBVDNA),得到约900bp的HBV-s基因片断。将其插入到噬菌粒载体pBluescriptSK+的Smal位点上。然后通过体外寡核昔酸介导的人工定点突变获得一系列(共12种)s基因“免疫逃逸”突变型。再通过EB病毒真核表达载体nMEP4将s基因突变型片断定向克隆到pMEP4上,从而构建了含HBV-s基因突变型的重组质粒pMEP4HBSM。用其转染人肝癌传代细胞系HepG2,经潮霉素选择,3周后获得抗性细胞克隆。经用HBV-s单克隆抗体检测发现除含变异体145(即145位上甘氨酸为精氨酸替代)外,其余变异体HBsAg均为阳性。经Westernblot杂交证实变异体145在分子量约为24000处也存有一特异性HBsAg蛋白带。这一表达细胞系的成功建立,为HBV免疫逃逸的诊断和治疗提供了基础。

To study the effection of overexpression of PML in growth and tumorigenicity of prostatic cancer cell, PSG5PML and SV2neo were co transfected into prostatic cancer cell(ET cell).The positive clone cells were selected by G418 and confirmed by Western blot.Then, using the in vitro cell growth assay and the in vivo tumorigenicity assay, the effection of overexpression of PML on growth and tumorigenicity of prostatic cancer cell was studied. Successful transfection of PML gene and effective expression of PML protein were confirmed by Western blot .The cells with overexpressed PML protein had a lower growth rate and a poorer tumorigenicity compared with control cells.The results demonstrated that overexpression of PML growth suppressor could suppress the growth and tumorigenicity of prostatic cancer cells in vitro and in vivo.

为探讨PML(Promyelocyticleakemia)生长抑制因子对人前列腺癌细胞的作用,采用lipofectamine介导基因转染技术将PML基因和G418抗性基因联合导入体外培养的人前列腺癌细胞,经G418筛选、Westernblot杂交检测出PML蛋白过表达细胞克隆,经体外生长抑制试验和裸鼠体内致瘤能力试验,观察PML过表达对前列腺癌细胞体外生长和裸鼠体内致瘤能力的影响。结果证实PML基因被成功地导入前列腺癌细胞中并得到高效表达。过表达PML蛋白的细胞体外生长能力和裸鼠体内致瘤能力明显低于对照细胞。提示PML生长抑制因子过表达能够有效地抑制前列腺癌细胞的体外生长和裸鼠体内的致瘤能力。

AIM To explore the effect of p27 gene on the growth inhibition of renal carcinoma cell line GRC 1.METHODS The p27 cDNA was transfected into human renal carcinoma cell line GRC 1 cells with lipofectamine. The growth rate, cell cycles and colony formation rate of transfected cells were observed by means of MTT and FCM assay. RESULTS Expression of p27 in GRC 1 was identified by Dot blot and Western blot analyses. The growth rate of GRC 1 transfected with p27 gene was markedly suppressed. Colony formation in soft agar was also decreased significantly. Cell cycle analysis by flow cytometry show that the number of cells in G 0 G 1 phase of GRC 1 cells was significantly increased while cells in S and G 2+M phase was decreased compared to that of the control GRC 1 cells. CONCLUSION Transduction of p27 gene into lower expression cancer cells can restore its suppressive effect on cell growth by arrest of cell cycle at G 1 phase.

目的 探讨 p2 7基因对肾癌 GRC- 1细胞系生长的抑制作用 ,为肾癌发生机制的研究和基因治疗提供理论依据 .方法 采用基因转染技术 ,将 p2 7c DNA转染到肾癌 GRC- 1细胞系中 ,了解其对细胞增殖能力及细胞周期的影响 .结果 转染 p2 7基因的肾癌细胞生长受到抑制 ,其在软琼脂上克隆形成能力降低了大约 4倍 .对细胞周期分析表明 ,处在 G0 -G1 期的细胞由 32 .2 %增加到 6 6 .9% ,经 Dot blot和 Westernblot杂交证实 ,p2 7m RNA表达有明显差异 (P<0 .0 1) ,p2 7的蛋白表达量有明显差异 (P<0 .0 1) .说明 p2 7蛋白具有生长抑制功能 ,其作用点为 G1 - S.结论 提高肾癌细胞中 p2 7的表达 ,能抑制肿瘤细胞的生长 ,导入 p2 7基因是一种治疗肾癌的新途径
