Thymocytes could be spontaneously fragmented (no treatment ) and occured during 8 hours of culture in vitro.

细胞凋亡(Apoptosis)发生在正常生理状态下的分化发育过程中,为胸腺内阴性选择的机制之一。本文报道分别将小鼠胸腺上皮细胞系MTEC1及其培养上清与裸鼠骨髓细胞进行共育,以染色体DNA发生规律性断裂为指标,对共育后的裸鼠骨髓细胞的凋亡状况进行分析,结果显示与MTEC1细胞共育后8小时,裸鼠骨髓细胞已发生凋亡,经细胞表型分析,凋亡的细胞可能为粒细胞和B细胞;而于DMEM中培养8小时以及与MTEC1上清共育8小时的裸鼠骨髓细胞末发生凋亡。结果表明MTEC1可主要通过细胞。细胞间相互作用诱导裸鼠骨髓细胞发生细胞凋亡,凋亡细胞可能主要为粒细胞和B细胞。

Results The results showed that when cells were cocultured with etoposide, cytosine arabinoside, vincristine or daunorubicine for 48 hours, APO% of normal bone marrow cells was 17.9%±4.6%, 30.5%±6.5%,...

DNA only from cells with cytosine arabinoside showed ladder. While APO% of HL 60 cells was 80.9%± 8.4% , 38.1% ±7.7%, 52.6%±9.3% and 24.2%±6.1%, respectively and sub G 1% was 73.4%±12.7%, 59.8%±10.1%, 47.6%±9.3% and 26.7%±6.8%, respectively. Ladder was seen in DNA from cells with the four drugs treatment. Conclusion These results demonstrated that normal bone marrow cells could be induced to apoptosis by chemotherapeutic agents. The effects of four drugs inducing apoptosis on HL 60 were stronger than those on normal bone marrow cells.

目的探讨化疗药物诱导正常骨髓及白血病细胞凋亡作用的差异。方法检测了临床常用4种药物作用后凋亡细胞出现的比例(形态学指标为APO%、流式细胞仪指标为sub-G1%)和细胞DNA断裂(ladder)的变化。结果足叶乙甙、阿糖胞苷、长春新碱、柔红霉素作用48小时,正常骨髓细胞APO%分别为17.9%±4.6%、30.5%±6.5%、16.7%±4.2%、12.3%±0.7%;sub-G1%分别为10.5%±3.1%、27.9%±4.7%、6.5%±1.5%、4.5%±1.5%、仅阿糖胞苷组有ladder出现。白血病细胞株APO%分别为80.9%±8.4%、38.1%±7.7%、52.6%±9.3%、24.2%±6.1%;sub-G1%分别为73.4%±12.7%、59.8%±10.1%、47.6%±9.3%、26.7%±6.8%,且都有ladder出现。结论化疗药物也能诱导正常骨髓细胞凋亡,但明显低于诱导白血病细胞凋亡的作用

Objective To search for the feature of apoptosis in bone marrow cells of patients with myelodysplastic syndromes (MDS) and investigate whether apoptosis is related to the pathogenesis of MDS. Methods Apoptosis of bone marrow mononuclear cells (BMMC) from 38 MDS patients was studied by TdT mediated dUTP nick end labeling (TUNEL) in situ, DNA ladder and shortterm BMMC culture in vitro. Results The average positive index (PI) in BMMC of 38 cases with MDS was 20.19%±11.07%. Significant differences were observed between MDS group and normal group ( P <0.001), as well as t AML, AML and IDA group ( P <0.01). Apoptosis of micromegakaryocytes was found with double labeling by TUNEL and APAAP of CD 41 . Follow up study on apoptosis of 10 patients with MDS showed that the PI in BMMC of the patients transforming from MDS to leukemia or having worse progression of illness decreased significantly. After 4 hour culture in vitro, BMMC of 3 MDS patients showed a significant increase in the average late stage apoptotic cell positive index (LPI) and an obvious DNA ladder. However, AML and IDA control groups did not show any small DNA fragments and high LPI. Conclusion It is suggested that there was an extensive apoptosis in BMMC of MDS. Apoptosis is related to MDS development and its conversion to leukemia. Examination of apoptosis in BMMC of MDS may provide valuble prognostic information.

目的探讨骨髓增生异常综合征(MDS)骨髓细胞凋亡特征及其病理学意义。方法用原位末端脱氧核糖核苷酸转移酶介导的dUTP缺口末端标记(TUNEL),DNA梯子和体外培养等方法对38例MDS患者骨髓单个核细胞(BMMC)凋亡进行研究。结果MDS患者BMMC的TUNEL凋亡细胞阳性指数(PI)为20.19%±11.07%,显著高于正常人、MDS转化的白血病、急性髓细胞白血病(AML)及缺铁性贫血(IDA)组,(P<0.001和P<0.01)。TUNEL和CD41碱性磷酸酶抗碱性磷酸酶免疫酶标法(APAAP)双标记证明MDS骨髓病态小巨核细胞发生了凋亡。动态观察10例MDS患者,转为白血病后凋亡细胞PI值显著下降,尚未转为白血病患者随病情恶化凋亡呈下降趋势。7例MDS患者BMMC体外培养后,晚期PI(LPI)显著增高,3例出现DNA梯子,AML和IDA组LPI未见明显升高,未出现DNA梯子。结论MDS骨髓细胞凋亡过度与演变和转为白血病有关。原位检测细胞凋亡可作为监测MDS预后指标
