An immunohistochemical method was used to identify the relationship between a target organ, its innervated preganglionic cell group and dopamine-beta-hydroxylase or neurophysin-1 fibers. After injection of True Blue into the adrenal gland of rat, frozen sections of the spinal cord were stained with antisera directed against DBH or NP-1. Under microscope with ultraviolet excitation, a small population of the sympathetic preganglionic neurons labeled by True Bluc are located in the lower thoracic segments. Turn to blue light excitation to examine the immunofluorescence active fibers, and using double exposure method to show the labeled cells and fluorescent fibers in one microphotograph. True Blue labeled cells receive abundant DBH fibers and few NP-1 fibers. That means the preganglionic cells which innervate the adrenal gland receive DBH fibers mainly. It suggests that a target organ may be innervated by a special group of preganglionic neurons, and in turn, they may receive specific neuronal input, characterized by a particular neurotransmitter.
