On the other hand,our results revealed that long term cryopreservation could reduce the antigen markers of OKT4 cells which was generally considered to be related to GVHD. It therefore seeilis reasonable to assume that multi-cord blood transfusion would not present serious problems. Thus two patients with advanced pelvic carcinoma were treated by intensive chemotherapy with HLA mismatched multi-cord blood transplantation. The hemopoiesis of bone marrow was reconstituted and the patients were roached in complete remission within 1 month following treatment. Our preliminary results were encouraging.

CD34+ cells were isolated from human umbilical cord blood by using a high-gradient magnetic cell sorting system with anti-CD34 monoclonal antibody coated with microbeads, and then inoculated onto a pre-formed irradiated bone marrow stroma to eval uate the grafting efficiency of CD34+ cells with rhGM-CSF or IL-3, alone or in combination. A short incubation of CD34+ cells with growth factors could rapidly reconstitute a long-term hemopoiesis in the long-term liquid culture system. This demonstrates that a brief treatment of CD34+ cells with GM-CSF or IL-3 can improve the grafting efficiency of transplanted umbilical cord blood cells.

利用抗CD34单克隆抗体吸附磁性微球的方法分离纯化脐带血CD34+细胞,将其种入照射后的成年骨髓基质。比较rhGM-CSF、IL-3及两者的联合对植入效率的促进作用。结果表明:经2h铺展贴壁后,对照组只有36%的CD34+细胞植入基质,而生长因子预处理组则有68—89.6%的CD34+细胞植入基质。在长期液体培养体系中则显示了植入CD34+细胞多的处理组造血重建快速而持久。表明GM-CSF和IL-3预处理将明显提高脐带血移植效率。
