为探讨小盾纤恙螨在EHFV传播中的媒介意义,观察了在该螨中EHFV经期传播的直接证据和能否在螨体内增殖。方法是将螨幼虫、若虫分批进行EHFV分离,测定病毒的TCID(50)/ml滴度。结果在该螨幼虫期和若虫期均分离到EHFV,但在幼虫演变到若虫前40天左右,EHFV可在其体内消失,以后再度出现;病毒滴度在幼虫期逐渐下降,随后再度上升。结果证明EHFV可经期传播,并在螨体内有增殖的现象。

Objective To study the impact of Leptotromhidium scutellaris as a vector in transmission of hemorrhagic fever. Methods Reverse transcription polymerase chain reaction (PCR) and in situ hybridization technique were used to detect RNA of hemorrhagic fever with renal syndrome virus (HFRSV). TCID 50 /ml of HFRSV were titrated for larva and nymph of chigger mites periodically. Results RNA of HFRSV was detected in the tissues of chigger mites, such as ovary cells, etc. by PCR and in situ hybridization....

Conclusion It provides direct evidence at molecular level for the role of chigger mite as a vector in transmission of HFRSV with theoretical and practical importance in prevention of HFRS.

目的用分子生物学方法研究小盾纤恙螨的媒介意义。方法聚合酶链反应和原位分子杂交法。结果从恙螨体内检测到肾综合征出血热病毒RNA;取恙螨幼虫、若虫定期做肾综合征出血热病毒TCID50/ml滴定,证明恙螨体内肾综合征出血热病毒可传播并有增殖现象;用原位分子杂交技术在恙螨的卵巢细胞、支肠细胞等组织中检测到肾综合征出血热病毒RNA。结论研究结果为恙螨的媒介意义提供了具有分子水平的直接证据,对肾综合征出血热的流行病学和预防具有重要的理论意义和实用价值。

Chigger mite had been studied as a vector by molecular biological methods since 1994.The results show that HFRSV-RNA could be detected by RT-PCR and Nested RT-PCR in chigger mite.TCID 50 /ml titer of HFRSV in larva and nymph detection demonstrated that HFRSV could be transmitted by trans-stadial and proliferated in chigger mites.HFRSV-RNA located in ovary cells and midgut cells of chigger mites.The above results indicated that chigger mite is a transmitting vector of HFRSV.

1997 年以来我们采用分子生物学方法对小盾纤恙螨的媒介意义进行研究。结果表明, 用 R T- P C R 和 Nested R T- P C R 从恙螨体内检测到 H F R Sr- R N A; 用原位分子杂交技术在恙螨的卵巢细胞、支肠细胞等组织中检测到 H F R S U- R N A。以上结果为恙螨的媒介意义提供了具有分子水平的直接证据, 对 H F R S 的流行病学和预防具有重要的理论意义和实用价值。
