Serum and liver biopsies or autopsies of 30 hepatitis B patients were studied for HBV DNA status. By spot hybridization, it was found that 9/17 ( 52.94% ) of HBsAg ( + )/HBeAg( + ) patients were serum HBV DNA positive, whereas in 2/6(33.33% ) of HBsAg ( + )/Anti-HBe ( + )pati-ents, serum HBV DNA was also detected. The specificity of the bands was checked by hybridization with pBR 322 probe, and 5/27 definitely showed high molecular weight hybridization bands with HBV DNA probe, which suggests HBV DNA integration. By using two Bio-11-dUTP labelled DNA probes specific for the Salmonella genus,i.e.,Bio-8.0 kb probe and Bio-3.4 kb probe,100 Salmonella isolates including 60 Salmonella serotypes were detected successfully by colony hybridization assays.In order to obtain the specific hybridization results,the colonies on nitrocellulose filter membrane required stringenttreatment with lysozyme,proteinase K,RNase and phenol-chloroform to effectively removeendogenous biotin,glycoproteins and structural analogs of biotin which produced nonspecific binding reactions with streptavidin.This study is useful to the general application of Salmonella DNA probe technique in basic level laboratory.

本实验应用Bio-11-dUTP标记的两个沙门氏菌属特异性核酸探针(Bio-8-0kb探针和Bio-3.4kb探针)成功地通过菌落杂交试验检测了60个血清型中的100株沙门氏菌。生长在硝酸纤维素滤膜上的菌落,经过溶菌酶,蛋白酶K、RNase及苯酚-氯仿严格的处理程序处理后,可以有效地消除内源性生物素、糖蛋白及生物素类似物等易与链亲和素结合产生非特异性反应的物质,从而获得了特异的杂交结果。此试验有利于使沙门氏菌核酸探针检测技术在基层得到推广应用。
