目的 研究血管内皮生长因子 (VEGF)受体flt 1胞外区基因抗肿瘤新生血管生成的作用。 方法 采用脂质体转染技术将含有flt 1胞外区前三个IgG样结构区域的真核表达质粒 pcD NA3.1/KDRn7转入人膀胱癌EJ细胞 ,用G418筛选得到稳定表达目的蛋白的细胞克隆 ,经固相结合实验筛选得到表达与VEGF特异结合的目的蛋白的细胞克隆 ,阳性细胞克隆进行PCR和RT PCR鉴定。对裸鼠人膀胱癌组织切片进行免疫组化染色 ,定量分析微血管密度。 结果 PCR和RT PCR结果显示重组质粒转入EJ细胞及在转录水平表达 ;免疫组化的微血管密度实验组明显低于阴性对照组。 结论 阻断VEGF/flt 1信号传导途径能够抑制肿瘤新生血管的形成 ,延缓肿瘤的生长速度

Objectives To measure the expression of E1AF and matrix metalloproteinase (MMP-2) on tissues of bladder transitional cell carcinoma(BTCC) and to discuss their relationship and clinical significance. Methods 41 human BTCCs were studied by RT-PCR and Western Blot to determine the expression of mRNA and protein of E1AF and MMP-2 respectively. After that, computer-assisted image scan and half-quantitative analysis were performed. Results In 26 superficial BTCCs, the positive rate of E1AF was 2690%, and the OD value of MMP-2 and E1AF was 3609±582 and 2908±626 respectively. In 15 invasive BTCCs, the positive rate of E1AF was 6667%, and the OD value of MMP-2 and E1AF was 11998±1044 and 8962±1096 respectively. Significant differences in the OD values of E1AF and MMP-2 and positive rate of E1AF were observed between the two groups. The expression of E1AF in grade Ⅲ was higher than that in grade Ⅰ. The expression of E1AF was significantly associated with that of MMP-2 in the bladder tissues (r=0536,P<001). Conclusion The expression of MMP-2 is regulated by E1AF. The high expression of E1AF and MMP-2 is associated with the invasion and prognosis of BTCC.

目的 测定人膀胱移行细胞癌组织中 ,不同病理分期的人细胞 5型腺病毒早期转录单位E1A因子 (E1AF)和基质金属蛋白酶 2 (MMP 2 )的表达 ,探讨二者之间关系及其临床意义。 方法 采用RT PCR和WesternBlot法 ,检测 41例人膀胱癌组织中E1AF和MMP 2的mRNA及蛋白表达 ,并运用计算机图像扫描分析仪测定图像的吸光度值 (A值 )进行半定量分析。 结果  2 6例表浅性膀胱癌中 ,E1AF的阳性率为 2 6 90 % ,MMP 2和E1AF的A值分别为 36 0 9± 5 82和 2 9 0 8± 6 2 6 ;15例浸润性膀胱癌中 ,E1AF的阳性率为 6 6 6 7% ,MMP 2和E1AF的A值分别为 119 98± 10 44和 89 6 2±10 96。E1AF的阳性率、MMP 2和E1AF的A值在两组均有显著性差异 ;E1AF在G3级膀胱癌组织中的表达量显著高于G1级 ;MMP 2和E1AF的表达呈正相关 (r=0 5 36 ,P <0 0 1)。 结论 E1AF调节MMP 2的表达 ;二者高表达与膀胱癌浸润有关
