Background/Aims: To evaluate the regulatory effect of calcium folinate (CF) on 5-fluorouracil (5-FU) against human gastric cancer cell. Methods: Primary cell culture technique and MTT colorimetric assay were used to study propagational inhibition effect of CF, 5-FU and a combination of the two drugs on 30 fresh human gastric cancer cell lines in vitro. Results: The immediate propagational inhibition rates of CF, 5-FU and CF/5-FU against gastric cell lines were 11.82%, 42.25% and 44.54% respectively. The combination...

Background/Aims: To evaluate the regulatory effect of calcium folinate (CF) on 5-fluorouracil (5-FU) against human gastric cancer cell. Methods: Primary cell culture technique and MTT colorimetric assay were used to study propagational inhibition effect of CF, 5-FU and a combination of the two drugs on 30 fresh human gastric cancer cell lines in vitro. Results: The immediate propagational inhibition rates of CF, 5-FU and CF/5-FU against gastric cell lines were 11.82%, 42.25% and 44.54% respectively. The combination of the two drugs had a slight rise in propagational inhibition rate in 43% of gastric cancer cell lines. Conclusions: CF increased the anticancer effect of 5-FU in small part of gastric cancer cell lines in vitro. The drug sensitivity test by MTT should be performed before chemotherapy using CF and 5-FU.

目的:探讨甲酰四氢叶酸钙(CF)调变5-氟脲嘧啶(5-FU)对不同胃癌原代细胞株的增殖抑制效应。方法:采用原代细胞培养和四唑氮蓝(MTT)药物敏感试验,观察大剂量CF相当于临床用药300mg/d)和5-FU(相当于临床用药750mg/d)单剂和联合应用对胃癌原代细胞的体外抑制作用。结果:体外对胃癌原代细胞株直接抑制作用,单剂CF为11.82%,单剂5-FU为42.25%;两者联合应用为44.54%,仅43%的胃癌原代细胞株显示CF对5-FU有一定的增效作用。结论:单剂CF对胃癌原代细胞仅有轻度抑制作用,联合应用大剂量CF和5-FU仅显示CF对部分细胞株5-FU的抑制作用有一定的增强效应,个体差异性较大,为确保CF的生化调节作用,避免其大剂量使用时毒副作用,建议用药前行MTT法药敏试验,以指导临床制订合理联合应用CF和5-FU的化疗方案。

Objective: To develop a kind of medicine that not only increases the anti-tumor activity but also decreases its toxicity by studying the biochemical modulation of green tea extract(TP 91). Methods: Models of mice transplantable solid carcinoma S180, and EAC were used. TP 91 and CTX were used. As the anticances drugs TP 91 and CTX were used during the experiment. Parameters of biochemical modulation, immunity and SOD were determined, and T test was used to analyse the results. Results: TP 91 possesses anti-tumor funcactivity. Its inhibition rate against S180 was 43.57%, and that of EAC was 40.15%. When TP 91 combined with CTX the inhibition rate against S180 mounts to 65.91%, and that against EACmounts to 61.36%. It also can be found that the coagulation time was lengthened that, the number of fibrinogen and the blood coagulabitity were reduced and that, erythrocyte count, SOD level as well as immune activity was increased. These results demonstrate that the TP 91 possesses an ability of biochemical modulation while inhibiting carcinoma.Conclusions: Acting as a kind of the anti tumor biochemical modulation agent, TP 91 is promising in cancer treatment and prevention. Further study is worth continuing.

目的 :通过绿茶提取物 (TP- 91)抗肿瘤生化调节研究 ,寻找既能增强抗肿瘤疗效 ,又降低其毒性的物质。方法 :采用小鼠 S180 ,EAC实体瘤为模型 ,以 TP- 91、CTX抗肿瘤实验并对血中生化、免疫、SOD等指标进行检测 ,所有数据以 t检验处理。结果 :TP- 91单用有抗肿瘤作用 ,S180抑瘤率 43.5 7% ,EAC抑瘤率 40 .15 %。而 CTX合并 TP- 91应用时 ,S180抑瘤率提高到 6 5 .91% ,EAC抑瘤率提高到 6 1.39%。实验中发现治疗组凝血时间延长 ,血浆纤维蛋白原下降 ,能改善血液高凝状态 ,提高免疫和红细胞 SOD活性增加 ,表现出 TP- 91在抗肿瘤中的生化调节作用。结论 :绿茶提取物 TP- 91,可作为抗癌生化调节剂之一 ,在肿瘤的治疗和预防中具有良好前景 ,值得深入研究。
