Studies in AML have suggested that some human acute myeloid leukemias arise from neoplastic transformation at the level of pluripotent stem cells while others arise at much later stages, leading to a block in cell maturation. In this study bone marrow and / or blood mononuclear cells from 11 patients with AML were observed in liquid suspension cultures. The results demonstrated that CPU-AML have two kinds of subpopulations. One grows exponentially in suspension cultures and forms colonies. The other grows exponentially in suspension, but does not form colonies even in cultures with methylcellulose, and has sufficient self-renewal capacity (exponential proliferation) for a long time. These cells could not form colonies of BFU-E, CFU-TL, CFU-GEMM and CFU-MK in the relevant culture systems. Cytochemical staining was negative, and monoclonal antibody fluorescent staining for lymphocytes was negative, too. These two subpopulations also have different characteristics in PHA dependence, morphology and survival time in vitro. We consider that these cells might arised at the level of leukemic stem cell.

目的探讨无精子不育症患者睾丸活检的临床价值及病变的组织学类型与预后的关系。方法分析104例无精子不育症患者睾丸活检标本的病理形态学特点。结果:104例无精子不育症中有101例为睾丸性不育(占9712%),其中以精子成熟停滞最多见。结论:通过对睾丸活检的病理观察,能对男性不育症的诊断、治疗措施的选择及预后的判断提供有益的依据。

Conclusion: Staining method may accurately distinguish spermatogenic cells from leukocytes,meanwhile, the morphological method of sperm cells also provide a new convenient method for learning fac-tors affecting spermatogenesis and the cause of infertile diseases.

目的:探讨精液生精细胞检测对不育症诊断的临床价值。方法:采用瑞-姬氏染色法,对生育组20例和不育组30例精液,进行生精细胞的检测与分类,并进行生精细胞定量计数。同时采用直接涂片法与染色镜检法对不育组30例精液进行白细胞检测对比。结果:两组绝大多数精液中都能看到精子细胞和精母细胞,而不育组70%的病例精原细胞的例数高于生育组(40%),表明不育组的多数病例存在生精障碍并导致生精细胞成熟停滞。两组生精细胞定量计数精母细胞与精子细胞,差异有显著性意义。用直接涂片法和染色镜检法两种方法检测白细胞,结果有显著性差异。结论:为提高精液检验的临床价值,须采用染色方法以鉴别生精细胞和白细胞。同时生精细胞的变化反映了生精功能的变化,根据生精细胞的类型可进一步衡量睾丸生精功能,为分析睾丸病因提供了客观指标。
