The same result was obtained by using dip method with diseased tissue.The electron microscopy of the ultrathin section of the chlorotic lesions incited by the virus infection on Chenopodium amaranticolor showed that there existed pinwheel-like inclusions in the cytoplasm of infected cells. The virus isolate was closely related, if not identical, to blackeye cowpea mosaic virus(BLCMV) serologically, and it also serologically related to, but distiuet from, cowpea aphidborne mosaic virus(CAMV), and bean common mosaic virus(BCMV). According to the results above, it is suggested that the seed-borne agent is blackeye cowpea mosaic virus(BLCMV).

1982年秋,在太安市效长豇豆田采集病株种子,以播种后所得的病菌为毒源,在苋色蔾上经单斑分离纯化,得一病毒分离物CV-sd。CV-sd侵染长豇豆“红嘴燕”品种所引起的症状为浓绿色脉带,花叶及叶片畸形。该分离物的寄主范围较窄,在测定的11个科的供试植物中,主要侵染豆科和蔾科植物。CV-sd的钝化温度为55°—60℃,稀释限点为10~(-2)。体外存活期为一天(室温),易经汁液传染和桃蚜,豆蚜、棉蚜以非持久方式传播,并经种子传播,其种传率为18.9%。电镜下观察,CV-sd质粒呈线条状,长720-740nm,电镜下观察病组织超薄切片,在受侵细胞质内存在大量风轮状内含体。用CV-sd提纯制品免疫家免所得的抗血清的效价为1/8。血清学测定结果表明,CV-sd与黑眼豇豆花叶病毒有密切的血清学关系,并与豇豆蚜传花叶病毒和菜豆普通花叶病毒有不同程度的血清学关系。据此,作者认为该分离物为黑眼豇豆花叶病毒。

从珊瑚豆(Solanum pseudocapsicum L.var diflorum(Vell)Bitter)花叶病株上分离到一病毒(SPV—I),汁液摩擦接种和桃蚜(Myzus persicea)、棉蚜(Aphis gossypii)均能传毒。两次试验证明该病毒不经种传,人工摩擦接种可侵染7科中的21种植物。在黄瓜、心叶烟等植物上能引起系统花叶,能引起苋色藜、昆诺阿藜接种叶片局部枯斑。该病毒钝化温度为65~70℃10分钟;稀释限点为10~(13)~10~(-4);体外存活期为1~2天(22℃)。琼脂免疫双扩散表明,此病毒与黄瓜花叶病毒有明显的血清学关系。浸蘸法制备样品,2%醋酸铀负梁,透射电镜观察,该病毒粒子为正二十面体,其直径为28~30nm。根据此病毒的生物学、血清学特性和病毒粒子的形态特征,初步认为该分离物是黄瓜花叶病毒的一个株系。
