The mathematical model of diffusion process in polarographic determination of dissolved oxygen has been thoroughly studied with an electric simulator. It is found that the combination of microelectrode and impulse operating technique is a possible way to improve the characteristics of present Clark electrode. The quantitative relations between the electrode size and operating condition are given in this paper in dimensionless form, and the range of the optimum condition has been discussed.

The respiratory inhibitor, NaF, of Embden-Me-yerhor-Parnas pathway (EMP) or the inhibitor, Na_3PO_4, of Hexose Monophatepathway (HMP) was added into the basal medium. Then calcoflur white VBLwas used to examine their effects on the formation of the cell wall. The uptakeof oxygen was measured with the electrode for dissolved oxygen....

Then calcoflur white VBLwas used to examine their effects on the formation of the cell wall. The uptakeof oxygen was measured with the electrode for dissolved oxygen. The results areas follows: 1. The regeneration of the cell wall of tobacco protoplasts is inhibited by10~(-3) M NaF,. Complete inhibition occurs at 10~(-2)--10~(-1) M (Fig. 1 A, B). 2. Inhibiting effect is also exercised by 10~(-3) M Na_3PO_4 on the regenerationof the cell wall. Inhibition is complete at 10~(-2)- 10~(-1) M (Fig. 1 C, D). 3. The percentage of inhibition of oxygen uptake by tobacco protoplasts is64.34% for 10~(-a) M NaF and 61.79% for 10~(-2) M Na_3PO_4 (Fig. 2A). 4. Oxygen uptake by tobacco protoplasts is more effectively inhibited by thecombined use of two inhibitors than by each used separately. Thus, the inhibi-tion percentage was 51.1 % for 5×10~(-4) M NaF+Na_3PO_4, while it was 31.81%for 10~(-a) M NaF, and 46.45% for 10~(-3) M Na_3PO_4 (Fig. 2B). 5. The inhibitory effect of 5×10~(-4) M NaF+Na_3PO_4 on cell wall rege-neration in tobacco protoplasts is greater than that of either 10~(-3) M NaF, or 10~(-3)M Na_3PO_4 alone. The percentages of inhibition were 42.0 %, 23.6 % and 12. 7%respectively (Fig. 2B).In light of the above findings, the perliminary assumption is that two path-ways, both EMP and HMP, are present and they may affect each other, and thusplay an important role in cell wall regeneration in tobacco protoplasts.
