Ber 10, 30 μmol/L reduced the percentage of cell death and LDH content in extra cellular bathing media in injuried cortical cultures. It also prevented the elevation of MDA and increased the activity of SOD and GSH Px. These results suggested that Ber could prevent the toxicity of NMDA and H 2O 2 or serum free culture insults by suppressing the generation of lipid peroxide and increasing the activities of antioxidant enzymes. 体外培养大鼠胎鼠大脑皮层神经细胞,观察小檗碱(berberine,Ber)对N-甲基-D-门冬氨酸(N-methyl-D-aspar-tate,NMDA)、过氧化氢(H2O2)及撤血清培养引起的细胞损伤的保护作用。结果表明,培养的大鼠胚胎大脑皮层神经细胞在NMDA500μmol/L作用10min、或H2O2100μmol/L24h、或无血清培养48h后,细胞的死亡率及乳酸脱氢酶(LDH)漏出率明显增加,抗氧化酶SOD、谷胱甘肽过氧化酶(GSH-Px)活性显著降低,而脂质过氧化物丙二醛(MDA)生成增多。Ber10、30μmol/L能显著降低细胞死亡率、LDH漏出率及MDA的生成,且能明显提高抗氧化酶活性。提示:Ber可拮抗NMDA、H2O2及撤血清培养引起的神经细胞损伤,其机理可能与其减少膜脂质过氧化物生成,提高抗氧化酶活性有关

Objective:To investigate the effect of serum starvation on expression of EGFR and the effect on expression of EGFR by EGF in human hepatocellular carcinoma cells.Methods:Human hepatocellular carcinoma cells were treated with serum free culture medium and the concentration of the 5ng/ml EGF,25ng/ml EGF in culture medium respectively for six hours,and then the expression of EGFR in BEL7402 human hepatocellular carcinoma cells was studyed by immunohistochemistry technique.Results:Our results indicate that the positive cells of EGFR expression in human hepatocellular carcinoma cells were(5.66±0.86)%、(10 67±1 26)% and (16.83±1.66)% respectively.Compared with normal control group(1.81±0.75)%,the EGFR expression of theirs was significantly higher than that of control group while the positive cells of the group treated with EGF were more higher than that of group cultured in serun free culture medium,and also the expression of EGFR of group treated with the concentration of the 25ng/ml EGF was higher than that of group treated with the concentration of the 5ng/ml EGF.Conclusion:Serum starvation and EGF can enhance the expression of EGFR in human hepatocellular carcinoma obviously.The up regulation of EGFR expression by EGF in HCC cells was showed dose depended.

目的 :探讨血清饥饿以及EGF对人肝癌细胞EGFR表达的影响。方法 :用免疫组化方法研究血清饥饿、5ng/mlEGF和 2 5ng/mlEGF对人肝癌细胞EGFR表达的影响。 结果 :撤血清培养组、5ng/mlEGF和 2 5ng/mlEGF刺激组人肝癌细胞EGFR的表达阳性细胞分别为 ( 5 66± 0 .86) %、( 10 67± 1.2 6) %和 ( 16 83± 1.66) % ,与正常对照组 ( 1 81± 0 .75) %相比较差异显著 (P <0 0 1) ,EGF刺激组比血清饥饿组明显增高 (P <0 0 1)、2 5ng/mlEGF组比 5ng/mlEGF组增高 (P <0 0 1)。 结论 :血清饥饿和EGF能明显增强人肝癌EGFR的表达 ,且EGF上调EGFR表达的作用具有剂量依赖效应。

为了对人硫氧还蛋白hTRX功能及其在休克治疗前景进行评价 ,将PCR扩增的hTRX基因连入pKPL 4载体 ,转化大肠杆菌pop2 136 .通过两步离子交换柱分离得到纯化的蛋白 ,进一步观察其稳定性及在体外对NFS 6 0和MCF 7细胞的促增殖活性 .结果发现 ,TRX蛋白可明显促进去细胞因子诱导的NFS 6 0细胞增殖和撤血清后MCF 7细胞的增殖 ,并发现外源性TRX可明显对小鼠内毒素血症起保护作用 .
